Disinfection by-products effect on swimmers oxidative stress and respiratory damage.

Disinfection by-products (DBPs) are generated through the reaction of chlorine with organic and inorganic matter in indoor swimming pools. Different DBPs are present in indoor swimming pools. This study evaluated the effects of different chlorinated formations in oxidative stress and lung damage in 20 swimmers after 40 min of aerobic swimming in 3 indoor pools with different characteristics. Biological samples were collected to measure lung damage (serum-surfactant-associated proteins A and B), oxidative stress parameters (plasma protein carbonylation and malondialdehyde, and whole-blood glutathione oxidation), and swimming exertion values (blood lactate) before and after exercise. Chemiluminescent method for detection of eutrophication sources by estimation of organic amino nitrogen and ammonium in water.

An automatic method has been developed for the estimation of organic amino nitrogen (CH2-NH) and ammonium in water samples. We propose a continuous flow system in which nitrogen compounds react with hypochlorite reagent to produce chloramines. Subsequently, the mixture is mixed with luminol, generating a chemiluminescence signal. The signal emission at 425 nm, registered as a function of time, decreases as nitrogen concentration increases, due to the decrease on hypochlorite concentration. A large number of nitrogen compounds have been assayed and their sensitivities compared, in milligrams per liter nitrogen. Abstract Due to the difficulties of working with chloramines, a critical examination of monochloramine standard preparation has been performed in order to select the best synthesis conditions. The analyte has been determined by in-tube solid phase extraction coupled to capillary liquid chromatography with UV detection (IT-SPME Capillary LC DAD). Potential factors affecting the response of monochloramine such as the pH of mobile phase and the volume of sample processed by IT-SPME Capillary LC DAD have been investigated and optimized. Selective flow-injection biamperometric determination of sulfur-containing amino acids and structurally related compounds

Abstract A simple and selective flow-injection method for the determination of cysteine and methionine is proposed. The method is based on the reaction of the amino acid with chloramine-T and the subsequent reaction of the excess of chloramine-T with an acidic solution of iodide producing a stoichiometric amount of tri-iodide (diminution of the flow-injection analysis (FIA) peak) which is determined biamperometrically using two platinum electrodes polarized at 100 mV. Flow injection biamperometric determination of chloramine-T in environmental, pharmaceutical and veterinary samples

Abstract A flow injection assembly for the determination of chloramine-T is proposed. The sample (213 μl) is inserted into the carrier, de-ionized water flowing at 4.1 ml min−1. This carrier merges with a mixture of potassium iodide and sulphuric acid, and the resulting solution flows to the flow cell through a reactor 66 cm long. The chloramine-T oxidises the iodide to tri-iodide. The resulting iodide/iodine ratio is biamperometrically tested. Cysteine-Specific Radioiodination of Proteins with Fluorescein Maleimide

A protocol is described for coupling of carrier-free iodine to protein sulfhydryl groups via fluorescein maleimide. 125I is first coupled to fluorescein maleimide in the presence of chloramine T. Iodination is stopped with sodium thiosulfate, and the iodine-substituted fluorescein maleimide is reacted with free cysteines of the protein. Excess label is then removed by gel-permeation chromatography. The procedure avoids exposition of the protein to oxidative conditions and does not require purification of the labeled carrier reagent. The multidrug resistance-associated protein 2 (MRP2/ABCC2) is involved in the efflux of endogenous and xenobiotic substrates, including several anticancer and antiviral drugs. The functional consequences of ABCC2 protein variants remain inconsistent, which may be due to shortcomings of the in vitro assays used. Differential effects of cysteine and methionine residues in the antioxidant activity of human serum albumin

Antioxidant properties of human serum albumin (HSA) may explain part of its beneficial role in various diseases related to free radical attack. In the present study, the antioxidant role of Cys and Met was studied by copper-mediated oxidation of human low density lipoproteins and by free radical-induced blood hemolysis which essentially assessed metal-chelating and free radical scavenging activities, respectively. Mild conditions were set up to specifically modify Cys and Met residues by N-ethylmaleimide (NEM) and chloramine T treatments, respectively.
