An in-tube SPME device for the selective determination of chlorophyll a in aquatic systems.

We report a new device for the estimation of the content of chlorophyll a pigment in water samples as an indicator of water quality. The extraction of the pigment from water was also optimized. 10 mL of water was filtered through a nylon filter (45m pore size and 13 mm of diameter), after the chlorophylls were dissolved by immersing the filter in 1 mL of a low non-hazardous solvent as ethanol. An in-valve in-tube SPME device coupled to capillary liquid chromatography with diode array detection was designed. Chlorophyll-Protein Complexes of Chlorella fusca

Chlorophyll-protein complexes from thylakoids of the normal type and two mutants of Chlorella fusca were separated using sodium dodecyl sulfate acrylamide gel electrophoresis (PAGE). The properties of the chlorophyll-protein complexes of the three strains of Chlorella were compared. Standard curves were set up for the characterization of the chlorophyll-proteins. In every electrophoretic separation of chlorophyll-protein complexes, a certain amount of pigment is separated from the protein. In photosynthesis the energy from the sun is captured by light harvesting chlorophyll pigments and converted to stable chemical energy, by the photochemical reaction center. The ligation of pigments to proteins involved in photosynthesis appears to be strictly regulated and, in turn, to have an important regulatory impact on the biogenesis of the photosynthetic apparatus. Even so, the molecular mechanism of pigment-protein assembly is largely unknown. The major light-harvesting chlorophyll a/b complex (LHCIIb) of photosystem II in higher plants can be reconstituted with pigments in lipid-detergent micelles. The pigment-protein complexes formed are functional in that they perform efficient internal energy transfer from chlorophyll b to chlorophyll a. LHCIIb formation in vitro, can be monitored by the appearance of energy transfer from chlorophyll b to chlorophyll a in time-resolved fluorescence measurements. Status of vulnerable Cystoseira populations along the Italian infralittoral fringe, and relationships with environmental and anthropogenic variables

We analyzed the occurrence and status of infralittoral fringe populations of Cystoseira spp. (Fucales) at thirteen rocky sites around the Italian coastline, and explored the relationships with relevant environmental and anthropogenic variables. We found Cystoseira populations at 11 sites: most were scattered and comprised monospecific stands of C. compressa, and only 6 sites also supported sparse specimens of either C. amentacea var. stricta or C. brachycarpa. Coastal human population density, Chlorophyll a seawater concentrations, sea surface temperature, annual range of sea surface temperature and wave fetch explained most of the variation of the status of C. compressa. Organic carbon and nitrogen stable isotopes in the intertidal sediments from the Yangtze Estuary, China

The natural isotopic compositions and C/N elemental ratios of sedimentary organic matter were determined in the intertidal flat of the Yangtze Estuary. The results showed that the ratios of carbon and nitrogen stable isotopes were respectively -29.8 per thousand to -26.0 per thousand and 1.6 per thousand-5.5 per thousand in the flood season (July), while they were -27.3 per thousand to -25.6 per thousand and 1.7 per thousand-7.8 per thousand in the dry season (February), respectively.
