Structural requirements for V2 vasopressin receptor proteolytic cleavage.

The ligand-induced proteolytic cleavage of the V2 vasopressin receptor transiently expressed in COS cells was investigated. After incubation of the cell membranes with a photoreactive ligand possessing full agonistic properties for V2 receptors, approximately 90% of the porcine and bovine V2 vasopressin receptors were cleaved in the upper part of transmembrane helix 2 at a heptapeptide sequence conserved in both vasopressin and oxytocin receptors. The oxytocin receptor was completely resistant to proteolysis after binding the same photoreactive ligand, which is only a partial agonist for this receptor. Neuroblastoma arises from neural crest cell precursors failing to complete the process of differentiation. Thus, agents helping tumor cells to differentiate into normal cells can represent a valid therapeutic strategy. Here, we evaluated whether guanosine (GUO), a natural purine nucleoside, which is able to induce differentiation of many cell types, may cause the differentiation of human neuroblastoma SH-SY5Y cells and the molecular mechanisms involved. This study examined whether a clinically relevant concentration of the volatile anaesthetic halothane modifies the endothelium-dependent relaxation produced by acetylcholine (3 nM-10 microM), histamine (1 pM-0.1 microM) and anti-human immunoglobulin E (1:1000) in human isolated pulmonary arteries submaximally precontracted with noradrenaline. An inhibitor of nitric oxide formation, N(G)-nitro-L-arginine (100 microM), attenuated acetylcholine-induced relaxation but failed to inhibit histamine- and anti-human immunoglobulin E-induced relaxation. Abstract In order to understand late magmatic processes that occur in the deep oceanic crust, we performed a phase-equilibria study in a representative late-stage system at a pressure of 200 MPa with a special focus on the role of water and oxygen fugacity. The starting composition for the experiments was evaluated based on a statistical approach using evolved fresh MORB glasses from the database PETDB highest in FeO and TiO2 (in average 17.92 wt% and 3.73 wt%, respectively), assumed to represent frozen liquids erupted at the seafloor generated by extensive differentiation of MORB. Mg-skarns enclosed in dunite cumulates of the Neo-Proterozoic Ioko-Dovyren intrusion (northern Baikal region, Russia) can be traced to silica-poor dolomitic host rock layers. The dominant minerals of the skarns are brucite (pseudomorph after periclase), forsterite and Cr-poor spinel. Rapid heating of quartzpoor dolomitic xenoliths led to the formation of minor olivine, followed by the breakdown of dolomite to calcite and periclase. Xenoliths were partially melted upon further heating resulting in a calcite melt. The mineralogically diverse eclogite xenolith suite sampled by the >1.6 Ga Zero kimberlite in the Kuruman group, close to the western edge of the Kaapvaal craton consists of bimineralic eclogites, orthopyroxene-bearing eclogites with and without sanidine, and kyanite eclogites. Garnet exsolutions in clinopyroxene occur in all Zero eclogite types, and orthopyroxene and, where present, sanidine are also exsolved from clinopyroxene in orthopyroxene-bearing eclogites. Mineral thermobarometry indicates that the last P–T condition prior to the kimberlite eruption was 900–950 °C at 3.5–4.0 GPa. Abstract This study focuses on the comenditic trachyte enclaves hosted in pantelleritic lava and pumices emplaced during one of the most recent eruptive events at Pantelleria (~ 6 ka). Enclaves range from mm-sized fragments to dm-sized blocks with spheroidal to amoeboid shapes and characteristic globular surfaces; they are crystal-rich with ~ 30 vol% large anorthoclase, less abundant Fe-rich olivine, clinopyroxene and Fe–Ti oxides. Vesicles ranging from a few mm to 1–2 cm in size are distributed throughout the enclave and are commonly filled with microlite-free vesicular glass.
