Gastrointestinal tumors: Phytochemical and drug combinations targeting the hallmarks of cancer
2021
Cancer is a worldwide burden resulting in millions of deaths each year. In particular, gastrointestinal tumors are life-threatening malignancies and one of the leading reasons for death in developed countries. Phytochemicals can be found in grains, vegetables, fruits and several foods. Many phytochemicals, such as curcumin, genistein, luteolin, vitexin-2-O-xyloside, avenanthramides, quercetin, epigallocatechin-3-gallate (EGCG), resveratrol, sulforaphane, piperine and thymoquinone have been used in combination with different chemotherapeutic agents for their synergistic anticancer effects against various forms of cancer. In this review, we describe the antitumor properties and biological eff…
Antisense gene therapy using anti-k-ras and antitelomerase oligonucleotides in colorectal cancer
2005
Aim: to test the efficacy of anti-k-ras and antitelomerase oligonucleotides for disabling colorectal cancer cell growth. Material and methods: an established human colorectal cancer cell line (SW 480, ATTC ® ) was used. Oligodeoxiribonucleotides (ODNs) have a phosphorotioate modification to ensure intracellular intake. We used an antitelomerase ODN (Telp5) and two anti-k-ras ODNs (AS-KRAS and ISIS). AS-KRAS is designed to join the k-ras oncogene’s exon 1. ISIS links to the terminal transcription unit 5’ of k-ras. Telp5 joins the template region of the hTR telomerase subunit. ODNs have been tested in different concentrations (1, 5, 10, 20 micromolar). Cell viability has been tested at 48 and…
Contemporary Outcomes for the Curative Treatment of Colorectal Cancer Pulmonary Metastases.
2023
Purpose: Treatment of pulmonary metastases (PM) from colorectal cancer (CRC) is the standard of care by several guidelines from Europe and the USA, but the validity of this strategy has been recently questioned, and the available evidence supporting this strategy is weak. We report the outcomes of a curative intent strategy in a very recent and homogenous series of patients. Methods: We did a retrospective review of all curative intent surgical or ablative treatment of PM from CRC performed consecutively in 3 French institutions from January 2015 to December 2019. Demographics, clinicopathological, and molecular characteristics were evaluated. Cox regression models were used to identify pro…
Analysis of the Thymidylate Synthase Gene Structure in Colorectal Cancer Patients and lts Possible Relation with the S-Fluorouracil Drug Response
2010
Thymidylate synthase (TS) catalyzes methylation of dUMP to dTMP and it is the target for the 5-Fluorouracil (5-FU) activity*. Barbour et al. showed that variant structural forms of TS in tumour cell lines confer resistance to fluoropyrimidines. We planned to perform the whole TS gene structure by means of sequencing techniques in human colorectal cancer (CRC) san-rples to try to identify the presence of any possible TS variant form that could be responsible of fluoropyrimidines drug resistance and of the worse prognosis. We performed the TS-DNA gene sequence in 68 CRC from patients of A, B, and C Dukes' stages and different histological grade, but we did not find any mutation in the TS-DNA …
Could a variant structural form of thymidylate synthase gene of metastatic colorectal cancer patients be related with the poor response to 5 Fu treat…
2008
Background: Thymidylate synthase (TS) catalyzes methylation of dUMP to dTMP and is the target of 5-fluorouracil (5-Fu). TS levels vary considerably among tumors and the response to 5-FU is influenced by the intratumoral activity of the enzyme, with high levels generally being associated with a poor response. Response to 5-FU also depends on TS structure and some researchers showed that variant structural forms of TS in tumour cell lines confer resistance to fluoropyrimidines so the aim our study is to analyze the whole coding regions of the TS gene to evaluate a possible mechanism of 5 Fu resistance. Materials and Methods: We performed the TS-DNA gene sequence in 68 colorectal cancer (CRC) …
Revisiting the disappearance of terrestrial dissolved organic matter in the ocean: a <i>δ</i>13&lt…
2014
Abstract. Organic carbon (OC) depleted in 13C is a widely used tracer for terrestrial organic matter (OM) in aquatic systems. Photochemical reactions can, however, change δ13C of dissolved organic carbon (DOC) when chromophoric, aromatic-rich terrestrial OC is selectively mineralized. We assessed the robustness of the δ13C signature of DOC (δ13CDOC) as a tracer for terrestrial OM by estimating its change during the photobleaching of chromophoric DOM (CDOM) from 10 large rivers. These rivers cumulatively account for approximately one-third of the world's freshwater discharge to the global ocean. Photobleaching of CDOM by simulated solar radiation was associated with the photochemical mineral…
Role of terrestrial carbon in aquatic UV exposure and photoprotective pigmentation of meiofauna in subarctic lakes
2015
Summary Aquatic organisms are adversely influenced by ultraviolet radiation (UV) and utilise photoprotective strategies, including pigmentation. We examined UV-protective melanin pigmentation of aquatic meiofauna (Cladocera) in relation to the UV exposure across 25 tree line lakes in Finland to address the potential effects of increased UV and altered input of UV-screening terrestrial dissolved organic carbon (DOC) on aquatic organisms. Bio-optical parameters, including concentration of DOC, the coloured dissolved organic matter (CDOM) fraction, a suite of carbon quality indices and chlorophyll a, were analysed from lake water, and their role in controlling underwater UV environment (measur…
Città, intonaci, colore. Problematiche inerenti al restauro degli intonaci storici
2012
Antiproliferative effects of bioaccessible fractions of honeys from Sicilian black honeybee (Apis mellifera ssp. sicula) on human colorectal carcinom…
2021
The aim of this study was to evaluate the antiproliferative activity of bioaccessible fractions (BFs) obtained by the internationally standardized INFOGEST static digestion method to Sicilian honeys of three distinct floral origins (Sulla, Thistle and Dill) and the Manuka honey (gold standard), and to compare their effects based on total polyphenol content (TPC). Differentiated CaCo-2 cells (intestinal-like) and non-differentiated CaCo-2 and HCT-116 colonic tumour-like cells were incubated for 24 h with BFs of honeys to test viability, apoptosis, mitochondrial membrane potential (MMP), ROS and cell cycle. TPC after digestion ranked in the following order: Dill > Thistle > Sulla > M…
High miR-196a levels promote the oncogenic phenotype of colorectal cancer cells.
2009
AIM: To analyze the relevance of the microRNA miR-196a for colorectal oncogenesis. METHODS: The impact of miR-196a on the restriction targets HoxA7, HoxB8, HoxC8 and HoxD8 was analyzed by reverse transcription polymerase chain reaction (RT-PCR) after transient transfection of SW480 cancer cells. The miR-196a transcription profile in colorectal cancer samples, mucosa samples and diverse cancer cell lines was quantified by RT-PCR. Transiently miR-196a-transfected colorectal cancer cells were used for diverse functional assays in vitro and for a xenograft lung metastasis model in vivo. RESULTS: HoxA7, HoxB8, HoxC8 and HoxD8 were restricted by miR-196a in a dose-dependent and gene-specific mann…
