Detection of primary DNA damage in Chlamydomonas reinhardtii by means of modified microgel electrophoresis.

1997

The assessment of genotoxic potential in surface water requires test methods, among which are those that detect initial DNA damage in organisms of aquatic biocenosis. The microgel electrophoresis (MGE) "comet assay" was applied to a ubiquitous unicellular green alga (Chlamydomonas reinhardtii) to detect DNA damage caused by genotoxins. For this, the test protocol described by Singh NP et al. [Exp Cell Res 175: 184-191, 1988] was modified. Major modifications were the use of alkaline lysis buffer with ionic detergents and the reduction of preincubation and electrophoresis times. Short-time exposure of Chlamydomonas to the well-known genotoxicants 4-nitroquinoline-1-oxide (4-NQO), N-nitrosodi…

A comparative investigation of DNA adducts, DNA strand breaks and gene mutations induced by benzo[a]pyrene and (+/-)-anti-benzo[a]pyrene-7,8-diol 9,1…

1997

Abstract Genotoxic effects of benzo[ a ]pyrene (BP) and its reactive metabolites (±)- anti -benzo[ a ]pyrene-7,8-diol 9,10-oxide ((±)- anti -BPDE) were comparatively investigated in vitro with the permanent human fibroblast cell line MRC5CV1. Induced DNA adducts were measured by 32 P-postlabeling, DNA strand breakage was determined by the comet assay and the HPRT gene mutation test was used to detect cytotoxicity and mutagenicity. Treatment of MRC5CV1 cells with S9 mix-activated BP or with (±)- anti -BPDE resulted in a concentration-dependent increase in DNA adducts and strand breaks. Genotoxic effects of BP and (±)- anti- BPDE were detected by 32 P-postlabeling and the comet assay with sim…

The Italian adaptation and validation of the Adult Sibling Relationship Questionnaire (ASRQ) in Italian emerging adults

2013

The aims of the present study were to provide a contribution to the validation of the Adult Sibling Relationship Questionnaire (ASRQ) (Stocker, Lanthier, & Furman, 1997) with a population of Italian emerging adults. The ASRQ measures individuals’ current perceptions of their own behaviour and feelings toward their sibling, as well as their perceptions of their sibling’s behaviour and feelings toward him or her by assessing fourteen fundamental dimensions (Intimacy; Affection; Emotional Support; Instrumental Support; Knowledge; Similarity; Admiration; Acceptance; Dominance; Competition; Antagonism; Quarrelling; Maternal Rivalry; and Paternal Rivalry), combined in order to form three macro-di…

Do viscosity and wettability of fluoride varnishes affect their fluoride release?

2020

BACKGROUND: There are several brands of fluoride varnishes in the market, but the dynamics of fluoride release from each one might be different. The purpose of this study was to evaluate in vitro the release of fluorides by fluoride varnishes and to determine the correlation with viscosity and wettability. MATERIAL AND METHODS: Forty four enamel blocks 5x5 mm were randomly divided into 4 groups (n=11) ((Duraphat®, Clinpro™ White Varnish, Flúor Protector® and control). We applied 30 milligrams of fluoride varnish to each specimen. The specimens were immersed in a Calcium Phosphate solution at a pH= 6.0. We evaluated the release of fluoride, by using a selective fluoride electrode, during 6 w…

Definition of a laboratory optimization protocol for road bitumen improved with recycled tire rubber

2012

Abstract Bearing in mind the need to answer one of the most frequent needs for civil construction, in general, and major project such as road infrastructures in particular, this paper presents the results of a laboratory experimental study in order to evaluate the performance improvement that is possible to achieve when modifying road bitumen with CRM (Crumb Rubber Modifier) from discarded tire rubber (TR), using a Wet Process. The environmental advantage is double, since the aim of the research is to obtain high performance road bitumen thanks to the re-use of a discarded material, such as the rubber tires, which otherwise would be dumped in scrap-yard. In particular, this experimental stu…

Swimming pool water--fractionation and genotoxicological characterization of organic constituents.

2005

Swimming pool water treatment in general includes flocculation, sand filtration, and subsequent disinfection with chlorine. The continuous chlorination and input of organic material by bathers in combination with recirculation of the pool water leads to an accumulation of disinfection by-products (DBPs) in the water. Several DBPs have been identified as human carcinogens and are thought to cause allergic asthma. Therefore, the elimination of DBPs is one major aim of pool water treatment. Using membrane filtration as an alternative treatment technology, DBPs can be removed more efficiently than with conventional treatment. In this study membrane filtration and genotoxicity testing were appli…

Editorial: Methodological issues in psychology and social sciences research

2023

In recent decades, the classical methods of studying the psychometric properties of tests have advanced to evolve into new innovative statistical methods, which allows obtaining a new view of the data. In this sense, progress has been made in the use of item response theory and in the use of structural equation models. Likewise, classical statistical inference methods have advanced in hypothesis testing and Bayesian statistics is rapidly making its way both in experimental studies and in psychometric studies. The aim of this issue has been to contribute to the dissemination of new research methodologies in quantitative and qualitative analysis in Psychology, as well as to evaluate the effec…

Measurement Invariance and Construct Validity of the Satisfaction With Life Scale (SWLS) in Community Volunteers in Vietnam.

2022

Worldwide, the Satisfaction With Life Scale (SWLS) has become the most widely used measure of life satisfaction. Recently, an authorized Vietnamese-language version has been introduced. Using a convenience sample comprising community volunteers from Ho Chi Minh City (N = 1073), confirmatory support was found for the cross-national constancy of the one-dimensional structure underlying the SWLS. Corrected item–total polyserial correlations and Omega coefficient were satisfactory. Using multi-group confirmatory factor analysis, configural, metric, and scalar invariance of the SWLS factorial structure were tested by gender, age, marital status, income, and educational level. Strong evidence of …

Viewing the Sun as an X-ray star

2003

The Sun is the late-type star we can study with the highest level of detail. In the interpretation of stellar data, therefore, it is often assumed that the physical processes of the coronae of late-type stars are similar to those of the solar corona, i.e. the "solar-stellar analogy". In order to investigate the validity of this assumption, we have started a program to study systematically the Sun as an X-ray star. Our program aims to explore how far the solar model can be applied to other stars. In this paper we review the results obtained from these studies and, in particular, we discuss the variability of a star identical to the Sun during its cycle, the contribution of different coronal …

DNA double-strand breaks trigger apoptosis in p53-deficient fibroblasts

2001

DNA double-strand breaks (DSBs) are induced by ionizing radiation (IR) and various radiomimetic agents directly, or indirectly as a consequence of DNA repair, recombination and replication of damaged DNA. They are ultimately involved in the generation of chromosomal aberrations and were reported to cause genomic instability, gene amplification and reproductive cell death. To address the question of whether DSBs act as a trigger of apoptosis, we induced DSBs by means of restriction enzyme electroporation and compared the effect with IR in mouse fibroblasts that differ in p53 status [wild-type (+/+) versus p53-deficient (-/-) cells]. We show that (i) electroporation of PVU:II is highly effici…
