Regulation of the hDlg/hScrib/Hugl-1 tumour suppressor complex.

2008

The proper function of the Scribble tumour suppressor complex is dependent upon the correct localisation of its components. Previously we observed dynamic relocalisation of the hDlg component under conditions of osmotic stress. We now show that the other two components of the complex, hScrib and Hugl-1 display similar patterns of expression. We demonstrate, by shRNA ablation of hScrib expression, that hDlg and Hugl-1 are in part dependent upon hScrib for their correct localization. However under conditions of osmotic stress this apparent dependency no longer exists: hDlg and Hugl-1 localise to cell membranes independently of hScrib. We also demonstrate an interaction between the three compo…

Simple, Fast, Bright, and Stable Light Sources

2012

In this work we show that solution-processed light-emitting electrochemical cells (LECs) based on only an ionic iridium complex and a small amount of ionic liquid exhibit exceptionally good performances when applying a pulsed current: sub-second turn-on times and almost constant high luminances (>600 cd m(-2) ) and power efficiencies over the first 600 h. This demonstrates the potential of LECs for applications in solid-state signage and lighting.

Potential anticancer heterometallic Fe-Au and Fe-Pd agents: Initial mechanistic insights

2013

A series of gold(III) and palladium(II) heterometallic complexes with new iminophosphorane ligands derived from ferrocenylphosphanes [{Cp-P(Ph2)═N-Ph}2Fe] (1), [{Cp-P(Ph2)═N-CH2-2-NC5H4}2Fe] (2), and [{Cp-P(Ph2)═N-CH2-2-NC5H4}Fe(Cp)] (3) have been synthesized and structurally characterized. Ligands 2 and 3 afford stable coordination complexes [AuCl2(3)]ClO4, [{AuCl2}2(2)](ClO4)2, [PdCl2(3)], and [{PdCl2}2(2)]. The complexes have been evaluated for their antiproliferative properties in human ovarian cancer cells sensitive and resistant to cisplatin (A2780S/R), in human breast cancer cells (MCF7) and in a nontumorigenic human embryonic kidney cell line (HEK-293T). The highly cytotoxic trimeta…

Phenoxyamidine Zn and Al Complexes: Synthesis, Characterization, and Use in the Ring-Opening Polymerization of Lactide

2019

International audience; Herein we report the synthesis of new ditopic ligands, which consist of a phenoxy group and N,N,N'trisubstituted amidines linked by a methylene spacer (L1-L4). Their coordination chemistry has been studied/investigated with Zn(II) and Al(III). Alkane elimination route between the phenol-amidine proligands (L1H-L4H) and Et2Zn led to dinuclear complexes [(L1-L4)ZnEt]2 (1a-4a) in which the Zn centers are chelated by phenoxyamidine ligands and bridged through the oxygen atom of the phenoxy groups. Salt metathesis reaction between two equivalents of the sodium amidine phenate L1Na and ZnCl2 led to a bis-chelate chiral spiro-complex (L12Zn) 1a'. Analogous alkane eliminatio…

Ubiquitin ligase Rsp5p is involved in the gene expression changes during nutrient limitation inSaccharomyces cerevisiae

2009

Rsp5p is an essential ubiquitin ligase involved in many different cellular events, including amino acid transporters degradation, transcription initiation and mRNA export. It plays important role in both stress resistance and adaptation to the change of nutrients. We have found that ubiquitination machinery is necessary for the correct induction of the stress response SPI1 gene at the entry of the stationary phase. SPI1 is a gene whose expression is regulated by the nutritional status of the cell and whose deletion causes hypersensitivity to various stresses, such as heat shock, alkaline stress and oxidative stress. Its regulation is mastered by Rsp5p, as mutations in this gene lead to a lo…

DNA Oxidation Photoinduced by Norharmane Rhenium(I) Polypyridyl Complexes: Effect of the Bidentate N,N′-Ligands on the Damage Profile

2018

Re(I)--polypyridyl complexes have interesting and distinctive photochemical and photosensitizing properties. This work describes the capability to induce (or photoinduce) DNA damage of three Re(I)-complexes with a naturally occurring alkaloid called norharmane (nHo) as ligand: [Re(CO)₃ (nHo)(L)]CF₃ SO₃ where L=2,2'-bipyridine (ReBpy), phenanthroline (RePhen) or dipyrido[3,2-a:2',3'-c]phenazine (ReDppz). The interaction of the complexes with DNA was investigated by steady-state and time-resolved spectroscopy. Data show that the mode and strength of interaction depend on the chemical structure of the bidentate ligand. The complexes show a major static contribution to the overall interaction, …

Oxovanadium(IV) complexes with [ONNO]-chelating ligands as catalysts for ethylene homo- and copolymerization

2014

Oxovanadium(IV) complexes with (ONNO)-type tetradentate Schiff base ligands: salen, acacen, aceten, acetph (H2salen = N,N'-ethylenebis(salicylideneimine), H2aceten = N,N'-ethylenebis(2-hydroxyacetophenoneimine), H2acacen = N,N'-ethylenebis(acetylacetonimine), H 2 acetph = N ,N ' -phenylene-1,2-b is (2-hydroxyacetophenoneimine)), were the first time investi- gated in ethylene polymerization and ethylene/1-octene copo- lymerization processes. In general, all these complexes are moderately active precatalyst for ethylene polymerization up- on activation with EtAlCl2 and they give high molecular weight linear polyethylenes. Their activity in copolymeriza- tion was found relatively low. However,…

Selective G-quadruplex stabilizers: Schiff-base metal complexes with anticancer activity

2014

The affinity of three square-planar nickel(II) (1), copper(II) (2) and zinc(II) (3) Schiff-base complexes for wild-type human telomeric (h-Telo) and protooncogene c-myc G-quadruplex (G4) DNA was investigated by UV-visible absorption spectroscopy and circular dichroism. DNA-binding constants (Kb) were determined by spectrophotometric titrations for both G4-DNA and B-DNA. The results obtained point out that the three metal complexes selectively bind G4-DNA with higher affinity, up to two orders of magnitude, with respect to B-DNA. The nickel(II) complex 1 was found to be the most effective G4-DNA stabilizer and the Kb values decrease in the order 1 > 2 ≈ 3. Innovative computational investigat…

Unprecedented reactivity of a Schiff base ligand in the co-ordination sphere of copper(I) complex towards β-diketones. Synthesis and X-ray characteri…

2002

International audience; The reactions of the copper(I) derivative [CuL(THF)(CH3CN)]PF6 (1), in which L is the di-imine/pyridine ligand 2,6-bis[1-(2,6-diisopropylphenylimino)ethyl]pyridine, with different β-diketones (2,4-pentanedione (acacH), 1,3-diphenyl-1,3-propanedione (dbm)) yielded the new Cu(I) salt [CuL′2]PF6 (2) (L′: 2-[acetyl]-6-[1-(2,6-diisopropylphenylimino)ethyl]pyridine). These reactions of β-diketones with a di-imine/pyridine ligand co-ordinated to a Cu(I) centre constitute the first example of a partial hydrolysis of such a Schiff base within the inner sphere of a Cu centre. (S)-2-[(2-Hydroxybenzyl)azaniumyl]-4-(methylsulfanyl)butanoate

2011

The zwitterionic title compound, C12H17NO3S, is a reduced Schiff base derived from (S)-N-(2-hydroxybenzylidene)methionine. An intramolecular interaction between the N—H and carboxylate groups forms a roughly planar (r.m.s. deviation = 0.1405 Å) five-membered ring containing the H(N), N, Cα, C(carboxylate) and O atoms in a pentagonal conformation. In the crystal, a supramolecular triangle-shaped motif is generated by molecules held together by O—H...O and N—H...O hydrogen bonds.
