Dual-targeting siRNAs.

2010

We have developed an algorithm for the prediction of dual-targeting short interfering RNAs (siRNAs) in which both strands are deliberately designed to separately target different mRNA transcripts with complete complementarity. An advantage of this approach versus the use of two separate duplexes is that only two strands, as opposed to four, are competing for entry into the RNA-induced silencing complex. We chose to design our dual-targeting siRNAs as Dicer substrate 25/27mer siRNAs, since design features resembling pre-microRNAs (miRNAs) can be introduced for Dicer processing. Seven different dual-targeting siRNAs targeting genes that are potential targets in cancer therapy have been develo…

Electron quantization in arbitrarily shaped Au islands on MgO thin films

2013

Low-temperature scanning tunneling microscopy has been employed to analyze the formation of quantum well states (QWS) in two-dimensional gold islands, containing between 50 and 200 atoms, on MgO thin films. The energy position and symmetry of the eigenstates are revealed from conductance spectroscopy and imaging. The majority of the QWS originates from overlapping Au 6p orbitals in the individual atoms and is unoccupied. Their characteristic is already reproduced with simple particle-in-a-box models that account for the symmetry of the islands (rectangular, triangular, or linear). However, better agreement is achieved when considering the true atomic structure of the aggregates via a densit…

A modal approach for the evaluation of the response sensitivity of structural systems subjected to non-stationary random processes

2005

A method for the evaluation of the response sensitivity of both classically and non-classically damped discrete linear structural systems under stochastic actions is presented. The proposed approach requires the following items: (a) a suitable modal expansion of the response; (b) the derivation in analytical form of the equations governing the evolution of the derivatives of the response (the so-called sensitivity equations) with respect to the parameters that define the structural model; (c) an extensive use of the Kronecker algebra for determining the analytical expressions of the sensitivity of the structural response statistics to non-stationary random input processes. Moreover, a step-…

A Multivariate Analysis of HIV-1 Protease Inhibitors and Resistance Induced by Mutation

2005

This paper describes the use of the multivariate statistical procedure principal component analysis as a tool to explore the inhibitory activity of classes of protease inhibitors (PIs) against HIV-1 viruses (wild type and more-frequent single mutants, V82A, V82F, and I84V) and against protease enzymes. The analysis of correlations between biological activity and molecular descriptors or similarity indexes allowed a reliable classification of the 51 derivatives considered in this study. The best results were obtained in the case of the I84V mutant for which a high number of predictions was achieved. On this basis, this statistical approach is proposed as a reliable method for the prediction …

Bezier curves approximation of triangularized surfaces using SVG

2006

This paper presents a technique to convert surfaces, obtained through a Data Dependent Triangulation, in Bezier Curves by using a Scalable Vector Graphics File format. The method starts from a Data Dependent Triangulation, traces a map of the boundaries present into the triangulation, using the characteristics of the triangles, then the estimated barycenters are connected, and a final conversion of the resulting polylines in curves is performed. After the curves have been estimated and closed the final representation is obtained by sorting the surfaces in a decreasing order. The proposed techniques have been compared with other raster to vector conversions in terms of perceptual quality.

CVBEM for solving De Saint-Venant solid under shear forces

2013

Abstract Evaluation of shear stresses distribution due to external shear forces applied to De Saint-Venant beams has been solved through Complex Variable Boundary Element Method properly extended, to benefit from advantages of this method, so far widely used for twisted solids. Extending the above method, further simplifications have been introduced such as those of performing line integrals only, instead of domain integrals. Numerical applications confirm accuracy and efficiency of the proposed extended version of the method, since the good agreement with results proposed in literature.

Next-Generation Sequencing in Clinical Practice

2019

Abstract During the past few decades, Sanger sequencing represented the “gold standard” technique. In order to better define the mutational status of several genes at the same time, next-generation sequencing methodologies have been introduced in the molecular laboratory workflow. In the era of personalized medicine, this technological improvement plays a key role in the comprehensive molecular characterization of cancer patients in order to get a “tile” target therapy. For different cancer patients, different target therapies are available and different genes serve as clinical it relevant biomarkers. This chapter reviews the principal features of these novel technologies and their applicat…

Regio- and stereoselective synthesis of spiro-heterocycles bearing the pyrazole scaffold via [3+2] cycloaddition reaction

2022

Abstract Herein we reported the utility of one-pot multicomponent based [3+2] cycloaddition reaction transformation to prepare a new two hybrids of spirooxindoles engrafted with pyrazole skeleton. Upon treatment of the electron-deficient olefins based pyrazole motif with in situ the generated azomethine ylides (AY) of sarcosine with the 6-chloro-isatin afforded spiroadducts. To enlighten the regio- and diastereo-selectivity of these spiroheterocycles, single crystal X-ray diffraction analysis was presented. Using Hirshfeld calculations, many short distance contacts such as O…H, Cl…H, N…H, H…C, C…C and Cl…S have a great impact on the molecular packing and the crystal stability of 8a and 8b. …
