Direct subphthalocyanine conjugation to bombesin vs. indirect conjugation to its lipidic nanocarrier

International audience; Bombesin (BBN) was covalently bound to graftable subphthalocyanine (SubPc) or to a cholesterol derivative, a component of a liposome that encapsulates non-graftable SubPc. The latter bioconjugation approach was suitable to address the stability of SubPc and was achieved by copper-free click-chemistry on the outer-face of the liposome. Liposomes were purified (FPLC) and then analyzed in size (outer diameter about 60 nm measured by DLS). In vitro binding studies allowed to determine the IC50 13.9 nM for one component of the liposome, cholesterol, conjugated to BBN. Polyamine conjugates of stigmasterol.

Abstract Three new polyamine conjugates with stigmasterol [(3β,22 E )-stigmasta-5,22-dien-3-ol] were synthesized and subjected to basic antimicrobial and cytotoxic tests. The conjugate derived from spermine, (3β,22 E )-stigmasta-5,22-dien-3-yl 4(12-amino-4,9-diaza-dodecylamino)-4-oxobutanoate ( 5c ), displayed considerable antimicrobial activity on Staphylococcus aureus at low concentration (50 μg mL −1 ). The cytotoxic activity was tested on cells of human T-lymfoblastic leukemia (IC 50  = 35.8 ± 10.3 μM ( 5c ) and IC 50  = 35.9 ± 5.7 μM ( 5b )) and normal human fibroblasts (IC 50  = 38.0 ± 2.8 μM ( 5c ) and IC 50  = 45.5 ± 1.9 μM ( 5b )). Conjugate 5a displayed no activity in both tests.

Some monomers, oligomers and polymers with conjugated triple bonds

In the first part new monoaminodiacetylenes with different numbers of CH2-groups, the formation of Perowskit-type Mn complexes and their polymerizability are described. Symmetrical diaminodiacetylenes and some derivatives therefrom have also been prepared. The second part deals with different types of polymers with diacetylene units in the backbone. Under irradiation color formation and crosslinking take place. Well-defined low molecular weight model compounds of polydiacetylenes is the subject of the third part. Finally rod-like but soluble polymers with alternating p-phenylene and acetylene units are described.

Synthesis of a MUC1-glycopeptide-BSA conjugate vaccine bearing the 3'-deoxy-3'-fluoro-Thomsen-Friedenreich antigen.

A novel MUC1-glycopeptide–BSA conjugate vaccine with a specifically fluorinated Thomsen–Friedenreich antigen side chain at Thr6 was prepared. Preliminary immunological experiments reveal specific binding of the tumor-associated glycopeptide antigen analog by anti-MUC1-mouse antibodies.

On the synthesis of functionalized porphyrins and porphyrin conjugates via β-aminoporphyrins

International audience; The synthesis of functionalized porphyrins and their conjugates from meso-tetraarylporphyrins through the acylation and the oxidation of beta-aminoporphyrins was investigated. 2,3-Dioxochlorins were prepared by the oxidation of a variety of beta-aminoporphyrins and subsequently used in a condensation reaction with functionalized aromatic aldehydes and ammonium acetate to form beta-functionalized porphyrins bearing a fused imidazole ring. Under optimized experimental conditions both reactions tolerate various functional groups and afford the products in an appropriate overall yield. Stimuli-responsive bile acid-based metallogels forming in aqueous media

Abstract The synthesis and gelation properties of a picolinic acid conjugated bile acid derivative in the presence of metal salts along with the stimuli-responsiveness of the systems are reported. The gels are formed in the presence of Cu 2+ ions in the solvent systems composed of 30–50% of organic solvent (MeOH, acetonitrile, or acetone) in water. The gels respond to various stimuli: they can be formed upon sonication or shaking, and their gel–sol transformation can be triggered by a variety of chemical species. NMR, MS, and SEM techniques are exploited in order to gain a deeper insight on the self-assembled systems.

Tetra-alkoxy substituted PPV derivatives: a new class of highly soluble liquid crystalline conjugated polymers

Two first examples of highly soluble tetra-alkoxy substituted PPV derivatives, poly(2,3,5,6-tetrahexyloxy-1,4-phenylene vinylene) (TH-PPV) and poly[2,3,5,6-tetra(2'-ethyl-hexyloxy)-1,4-phenylene vinylene] TEH-PPV, are presented. Both polymers have been fully characterized and the solubility and UV-Vis absorption characteristics have been studied in various organic solvents. Due to the symmetric nature of the repeating unit, TH-PPV and TEH-PPV have an inherently regioregular nature, which is unusual for PPV-type polymers. Origin of the Red Sites and Energy Transfer Rates in Single MEH-PPV Chains at Low Temperature

Single poly[2-methoxy-5-(2'-ethyl-hexyloxy)-1,4-phenylene vinylene] (MEH-PPV) molecules dispersed in thin poly(methylmethacrylate) films have been investigated by fluorescence emission, excitation and time-resolved spectroscopy at 1.2 K. For the molecular weight studied (∼200 kDa) a bimodal distribution of emission maxima is observed. Based on a comparison of the spectroscopic properties of blue and red sites and on polarisation-resolved measurements, we argue in agreement with recent quantum-chemical calculations that the red subpopulation most probably does not arise from interchromophoric excitation delocalisation but is to be attributed to longer chromophoric units originating from orde…

Second order optimality conditions with applications

International audience; The aim of this article is to present the algorithm to compute the first conjugate point along a smooth extremal curve. Under generic assump- tions, the tra jectory ceases to be optimal at such a point. An implementation of this algorithm, called cotcot, is available online and based on recent devel- opments in geometric optimal control. It is applied to analyze the averaged optimal transfer of a satellite between elliptic orbits.
