Synthesis and biopharmaceutical characterisation of new poly(hydroxyethylaspartamide) copolymers as drug carriers.

2001

Abstract Four new poly(hydroxyethylaspartamide)-based copolymers bearing (a) poly(ethylene glycol) 2000, (b) poly(ethylene glycol) 5000, (c) poly(ethylene glycol) 2000 and hexadecylalkyl, (d) poly(ethylene glycol) 5000 and hexadecylalkyle, as pendant groups were synthesised. The copolymers were obtained by partial aminolysis of polysuccinimide with poly(ethylene glycol) and hexadecylalkyl amino derivatives followed by reaction with ethanolamine. Naked polyhydroxyaspartamide was obtained by polysuccinimide reaction with ethanolamine. The nuclear magnetic resonance, infrared, light scattering and elemental analysis allowed for the extensive physico-chemical characterisation of the carriers. T…

Soluble oligoaramide precursors--a novel class of building blocks for rod-coil architectures.

2005

A new synthetic route is described that allows the reversible conversion of the inherently insoluble oligo-p-benzamides into soluble materials through the formation of imidoyl chlorides. Syntheses of the corresponding dimer, trimer, and tetramer are reported; these compounds can easily be purified by crystallization and are accessible on the multigram scale. Structural proof was obtained by single-crystal X-ray structures of the trimer and tetramer precursors. They can be selectively functionalized into amides or esters at the terminal carboxylic acid group followed by hydrolysis of the imidoyl chlorides to the parent amides. This new class of compounds gives access to strongly aggregating …

Controlling the Polymer Microstructure in Anionic Polymerization by Compartmentalization.

2018

An ideal random anionic copolymerization is forced to produce gradient structures by physical separation of two monomers in emulsion compartments. One monomer (M) is preferably soluble in the droplets, while the other one (D) prefers the continuous phase of a DMSO-in-cyclohexane emulsion. The living anionic copolymerization of two activated aziridines is thus confined to the DMSO compartments as polymerization occurs selectively in the droplets. Dilution of the continuous phase adjusts the local concentration of monomer D in the droplets and thus the gradient of the resulting copolymer. The copolymerizations in emulsion are monitored by real-time 1 H NMR kinetics, proving a change of the re…

Redox-Responsive Block Copolymers: Poly(vinylferrocene)-b-poly(lactide) Diblock and Miktoarm Star Polymers and Their Behavior in Solution

2013

The synthesis of diblock and miktoarm star polymers containing poly(vinylferrocene) (PVFc) and poly(l-lactide) (PLA) blocks is introduced. End functionalization of PVFc was carried out via end capping of living carbanionic PVFc chains with benzyl glycidyl ether (BGE). By hydrogenolysis of the benzyl protecting group a dihydroxyl end-functionalized PVFc was obtained. Both monohydroxyl- and dihydroxyl-functionalized PVFcs have been utilized as macroinitiators for the subsequent polymerization of l-lactide via catalytic ring-opening polymerization. A series of block copolymers and AB2 miktoarm star polymers was synthesized with varied PLA chain lengths. All polymers were characterized in detai…

A minimal hydrophobicity is needed to employ amphiphilic p(HPMA)-co-p(LMA) random copolymers in membrane research.

2014

Because a polymer environment might be milder than a detergent micelle, amphiphilic polymers have attracted attention as alternatives to detergents in membrane biochemistry. The polymer poly[N-(2-hydroxypropyl)-methacrylamid] [p(HPMA)] has recently been modified with hydrophobic lauryl methacrylate (LMA) moieties, resulting in the synthesis of amphiphilic p(HPMA)-co-p(LMA) polymers. p(HPMA)-co-p(LMA) polymers with a LMA content of 5 or 15% have unstable hydrophobic cores. This, on one hand, promotes interactions of the hydrophobic LMA moieties with membranes, resulting in membrane rupture, but at the same time prevents formation of a hydrophobic, membrane mimetic environment that is suffici…

Thermodynamics of copolymer solutions: how the pair interactions contribute to the overall effect.

2014

Vapor pressure measurements were performed for solutions of poly(methyl methacrylate-ran-tert-butyl methacrylate) with different weight fractions of tert-butyl methacrylate units, and their parental homopolymers in chloroform at 323 K, over a large domain of concentrations. The Flory–Huggins interaction parameters obtained from these experimental investigations show complex dependences of the Flory–Huggins interaction parameter on concentration and copolymer composition. This behavior can be modeled by taking into account an approach which considers the ability of the polymers to rearrange in a response to changes in their molecular surroundings [Adv. Polym. Sci. 2011, 238, 1–66]. According…

Biocompatible polymeric micelles with polysorbate 80 for use in brain targeting.

2008

In this paper, the synthesis and characterization of novel amphiphilic graft copolymers based on an alpha,beta-poly(N-2-hydroxyethyl)-D, L-aspartamide (PHEA) backbone and D, L-polylactic acid (PLA) hydrophobic side chains are reported. These copolymers were obtained starting from PHEA-ethylenediamine (PHEA-EDA), which was functionalized with polysorbate 80 (PS(80)) and/or PLA in order to obtain the PHEA-EDA-PS(80)-PLA and PHEA-EDA-PLA samples, respectively. The degrees of derivatization, DD(PS80) and DD(PLA), of PHEA-EDA-PS80-PLA, calculated by (1)H-NMR, resulted in being 1.2 +/- 0.03 mol% and 0.54 +/- 0.05 mol%, respectively, while that of PHEA-EDA-PLA was found to be 0.60 +/- 0.05 mol%. Effect of polystyrene coating on pore, structural and chromatographic properties of silica packings

1990

Abstract A series of mesoporous and macroporous silicas with a mean pore diameter (PD), between 6 and 200 nm were reacted with a copolymer of styrene and vinyl-methyldiethoxysilane. The mass load of coated silicas corresponded to the monolayer capacity of the polymer calculated on the basis of the molecular cross-sectional area, except for the two mesoporous silcias of PD = 6 and 7.5 nm. Depending on the pore size, changes occurred in the specific surface area, as, between the native and the coated product. A comparison of the specific pore volume, νp, of silicas before and after polymer immobilization indicated a major loss of νp for the two mesoporous silicas whereas this effect was much …

Interfacial tension between polymer-containing liquids - Predictability and influences of additives

1999

The first part of the contribution deals with the interfacial tension, σ, of phase-separated polymer solutions in single or mixed solvents and of binary polymer blends as a function of the relative distance to the critical temperature of the system, special attention being paid to the possibilities of theoretical prediction. Two methods are discussed in more detail. One is based on a realistic description of the Gibbs energy of mixing as a function of composition, the second correlates σ with the length of the measured tie line. The second part is devoted to another aspect, namely the effects of additives on the interfacial tension between the coexisting phases of demixed polymer solutions …
