Exploiting Cryo-EM Structural Information and All-Atom Simulations To Decrypt the Molecular Mechanism of Splicing Modulators.

Splicing modulators (SMs) pladienolides, herboxidienes, and spliceostatins exert their antitumor activity by altering the ability of SF3B1 and PHF5A proteins, components of SF3b splicing factor, to recognize distinct intron branching point sequences, thus finely calibrating constitutive/alternative/aberrant splicing of pre-mRNA. Here, by exploiting structural information obtained from cryo-EM data, and by performing multiple μs-long all-atom simulations of SF3b in apo form and in complex with selected SMs, we disclose how these latter seep into the narrow slit at the SF3B1/PHF5A protein interface. Structural effects of macrocyclic compounds and their partition in sodium dodecylsulphate aqueous solutions

The partition of 1,4,7,10,13,16-esaoxacyclooctadecane (18C6), 4,7,13,16-tetraoxa-1,10-diazacyclooctadecane (2.2), 2,5,8,11,14,17-esaoxabicyclo[16.4.0]dicosane (B18C6) and 2,5,8,15,18,21-esoxatricyclo[20.4.0.09.14]esacosane (Cy218C6) in sodium dodecyl sulfate (SDS) aqueous solutions and their effect on the structure of surfactant aggregates has been investigated by small-angle neutron scattering. Results from data analysis have shown that by increasing macrocycle concentration the SDS micelles dimensions reduce for all systems investigated. Next-Day Bitcoin Price Forecast

This study analyzes forecasts of Bitcoin price using the autoregressive integrated moving average (ARIMA) and neural network autoregression (NNAR) models. Employing the static forecast approach, we forecast next-day Bitcoin price both with and without re-estimation of the forecast model for each step. For cross-validation of forecast results, we consider two different training and test samples. In the first training-sample, NNAR performs better than ARIMA, while ARIMA outperforms NNAR in the second training-sample.
