The quartz crystal microbalance (QCM) technique is an emerging bioanalytical tool to study the behavior of animal cells in vitro. Due to the high interfacial sensitivity of thickness shear mode (TSM) resonators it is possible to monitor the formation and breakage of cell-matrix interactions and changes in viscoelasticity of the cell bodies, as well as minute cell volume alterations by the time course of their resonance frequency even with millisecond time resolution. AbstractHuman papillomavirus type 33 (HPV-33) pseudovirus infection is a slow process dependent on the initial interaction with cell-surface heparan sulfate (T. Giroglou, L. Florin, F. Schafer, R. E. Streeck, and M. Sapp, 2001a, J. Virol. 75, 1565–1570). We have now further dissected the initial steps of pseudovirus uptake using removal of cell-surface proteoglycans and selective inhibition of entry pathways. Inhibition of Receptor Signaling to Phospholipase D by Clostridium difficile Toxin B

Rho proteins have been reported to activate phospholipase D (PLD) in in vitro preparations. To examine the role of Rho proteins in receptor signaling to PLD, we studied the effect of Clostridium difficile toxin B, which glucosylates Rho proteins, on the regulation of PLD activity in human embryonic kidney (HEK) cells stably expressing the m3 muscarinic acetylcholine receptor (mAChR). Reconstitution of vesicular transport to Rab11-positive recycling endosomes in vitro.

Rab GTPases are key regulators of vesicular protein transport in both the endocytic and exocytic pathways. In endocytosis and recycling, Rab11 plays a role in receptor recycling to plasma membrane via the pericentriolar recycling compartment. However, little is known about the molecular requirements and partners that promote transport through Rab11-positive recycling endosomes. ABSTRACT Virus-induced alterations in cell morphology play important roles in the viral life cycle. To examine the intracellular events of coxsackievirus B3 (CVB3) infection, green monkey kidney (GMK) cells were either inoculated with the virus or transfected with the viral RNA. Various microscopic and flow cytometric approaches demonstrated the emergence of CVB3 capsid proteins at 8 h posttransfection, followed by morphological transformation of the cells. Clustering transmembrane-agrin induces filopodia-like processes on axons and dendrites

The transmembrane form of agrin (TM-agrin) is primarily expressed in the CNS, particularly on neurites. To analyze its function, we clustered TM-agrin on neurons using anti-agrin antibodies. On axons from the chick CNS and PNS as well as on axons and dendrites from mouse hippocampal neurons anti-agrin antibodies induced the dose- and time-dependent formation of numerous filopodia-like processes. The processes appeared within minutes after antibody addition and contained a complex cytoskeleton. Secretion of Protein-bound Hydroxyproline from Moss Callus Cells

Abstract A glycoprotein rich in hydroxyproline was found in wall preparations of callus cells of the moss Physcomitrium pyriforme Brid. It is apparently attached to the non-cellulosic polysaccharides of the wall, and the majority is extractable by boiling the wall fraction or by using a chaotropic salt at room temperature. A pulse-chase technique was used to study the transport of this protein to the wall. Cytochalasin B seems to inhibit its secretion from the callus cells.
