Avidin Is a Promising Tag for Fusion Proteins Produced in Baculovirus-Infected Insect Cells

The baculovirus expression vector system (BEVS) has become one of the most versatile and powerful eukaryotic systems for recombinant protein expression. We have constructed a novel baculovirus transfer vector (pbacAVs+C) which allows for the efficient production, detection, and single-step purification of the desired molecule as a secretion-compatible avidin fusion protein in insect cells. It also enables fast construction of the baculoviruses by site-specific transposition in Escherichia coli. Diverse compounds mimic Alzheimer disease–causing mutations by augmenting Aβ42 production

Increased Abeta42 production has been linked to the development of Alzheimer disease. We now identify a number of compounds that raise Abeta42. Among the more potent Abeta42-raising agents identified are fenofibrate, an antilipidemic agent, and celecoxib, a COX-2-selective NSAID. Many COX-2-selective NSAIDs tested raised Abeta42, including multiple COX-2-selective derivatives of two Abeta42-lowering NSAIDs. Compounds devoid of COX activity and the endogenous isoprenoids FPP and GGPP also raised Abeta42. Introduction Rat liver epoxide hydratase [EC 4.2.1.631 which catalyses the conversion of epoxides to trurans-dihydro- diols has been purified to apparent homogeneity as determined by three independent criteria [l] . The preparation obtained was capable of catalysing the hydration of both styrene oxide and the 4,5- (K- region)epoxide of benzo(a)pyrene [ 11. Epoxides of polycyclic hydrocarbons have been implicated as the agents responsible for the cytotoxic and carcinogenic properties of such compounds (for reviews see [2-41). Direct comparison of the amino acid sequences of microsomal and soluble epoxide hydrolase superficially indicates that these enzymes are unrelated. Both proteins, however, share significant sequence similarity to a bacterial haloalkane dehalogenase that has earlier been shown to belong to the alpha/beta hydrolase fold family of enzymes. The catalytic mechanism for the dehalogenase has been elucidated in detail [Verschueren et al. (1993) Nature 363, 693-698] and proceeds via an ester intermediate where the substrate is covalently bound to the enzyme. Dynamic evaluation of aggregation and agglutination of red blood cells.

Viscoelastic evaluation of aggregation and agglutination of red blood cells was attempted. A concentric double cylinder viscoelastometer was used for determining the dynamic rigidity modulus and loss modulus of blood sample. The dynamic rigidity modulus of horse blood were measured over a wide range of hematocrit. The relation between the viscoelastic behavior and the erythrocyte sedimentation was examined. The change in the amount of surface charge of enzyme treated red blood cells was qualitatively estimated from the measurements of dynamic viscoelasticity of red blood cells suspension with added poly-L-lysine. Phenoloxidase-dependent cytotoxic mechanism in ascidian (Styela plicata) hemocytes active against erythrocytes and K562 tumor cells.

The cytotoxic activity against rabbit erythrocytes (RE) and human K562 tumor cells by Styela plicata hemocytes was significantly related to the phenoloxidase (PO) which converts phenols to quinone and initiates the melanogenic pathway. The effector hemocyte population, separated in a Percoll density gradient band, enriched in a granulocyte type named "morula cells", was examined with RE in a hemocyte cytotoxic assay and plaque forming cell assay. ABSTRACT Photobacterium damselae subsp. damselae is a marine bacterium that causes septicemia in marine animals and in humans. Previously, we had determined a major role of pPHDD1 plasmid-encoded Dly (damselysin) and HlyA (HlyA pl ) and the chromosome-encoded HlyA (HlyA ch ) hemolysins in virulence. However, the mechanisms by which these toxins are secreted remain unknown. In this study, we found that a mini-Tn 10 transposon mutant in a plasmidless strain showing an impaired hemolytic phenotype contained an insertion in epsL , a component of a type II secretion system (T2SS). Changes of 1-Aminocyclopropane-1-Carboxylic Acid Oxidase Activity in Stressed Pinus Sylvestris Needles

Stimulation of ethylene biosynthesis in pine needles by hydrogen peroxide and sodium bisulfite coincided with the activation of ACC oxidase at the level of protein synthesis. Decrease in ethylene production at high concentrations of sodium bisulfite (above 7 mM) was apparently due to inhibition of ACC oxidase activity. Treatment of pine needles with aminotriazole caused an inhibition of both ethylene production and ACC oxidase activity. Both methylviologen and methyl jasmonate stimulated ACC oxidase activity in a concentration-dependent manner with no parallel changes in ethylene production. Summary The main objective of the present study was to test the hypothesis that increased concentrations of ethylene under conditions of limited gas exchange during in vitro cultivation of potato ( Solanum tuberosum L.) stem explants may lead to increased capaciry of antioxidative defense.
