Partial T Cell-Depleted Peripheral Blood Stem Cell Transplantation from HLA-Identical Sibling Donors for Patients with Severe Aplastic Anemia

2019

We analyzed the outcomes of 26 consecutive patients with acquired severe aplastic anemia (SAA) undergoing peripheral blood stem cell transplantation (PBSCT) with partial ex vivo T cell depletion with a targeted T cell dose from HLA-identical sibling donors. The median patient age was 37 years (range, 3 to 63 years). Four patients with uncontrolled pneumonia at the time of transplantation died, on days +1, +2, +21, and +26. All evaluable patients engrafted, with a median time to neutrophil recovery of 11 days (range, 10 to 14 days) and a median time to platelet recovery of 19 days (range, 8 to 53 days). Two patients had transient grade I acute graft-versus-host disease (GVHD) with skin invol…

Depletion of Alloreactive Donor T Lymphocytes by CD95-Mediated Activation-Induced Cell Death Retains Antileukemic, Antiviral, and Immunoregulatory T …

2007

In allogeneic hematopoietic stem cell transplantation (AHSCT) graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) effect are closely but not invariably linked. Thus, harnessing donor lymphocyte mediated GVL immunity and separating it from GVHD is of particular interest. Based on results obtained in murine models we have explored the CD95-mediated activation-induced cell death (AICD) strategy to selectively deplete alloreactivity in human donor T lymphocytes in vitro. Following stimulation of CD3(+) T cells isolated from HLA-A* 0201-positive donors with HLA or minor histocompatibility antigen mismatched hematopoietic or nonhematopoietic cells in the presence of agonistic anti-CD…

Geochemical processes assessed by Rare Earth Elements fractionation at "Laguna Verde" acidic-sulphate crater lake (Azufral volcano, Colombia)

2017

Abstract The geochemical behaviour of major elements, Fe, Al, Mn, and Rare Earth Elements (REE) was investigated in the “Laguna Verde” acidic crater lake of Azufral volcano (Colombia). The cold lake water (T close to 10 °C) is sulphate-dominated, due to absorption and oxidation of H2S (pH 2.1–2.7, Eh 196–260 mV), and Na-enriched (Total Dissolved Solids 0.79 g L−1). The total amount of REE dissolved in the lake ranges from 3.3 to 9.1 ppb. The REE patterns normalized to the local rocks show a Light Rare Earth Elements (LREE) depletion quite constant in the 15 samples. Similar patterns were already found in the acidic sulphate springs of Nevado del Ruiz volcano-hydrothermal system, caused by t…

Depletion of alloreactive T cells via CD69: implications on antiviral, antileukemic and immunoregulatory T lymphocytes

2005

Selective depletion of alloreactive T cells from stem-cell allografts should abrogate graft-versus-host disease while preserving beneficial T cell specificities to facilitate engraftment and immune reconstitution. We therefore explored a refined immunomagnetic separation strategy to effectively deplete alloreactive donor lymphocytes expressing the activation antigen CD69 upon stimulation, and examined the retainment of antiviral, antileukemic, and immunoregulatory T cells. In addition to the CD69high T cell fraction, our studies retrieved two T cell subsets based on residual CD69 expression. Whereas, truly CD69(neg) cells were devoid of detectable alloresponses to original stimulators, CD69…

Tumor Blood Flow and O2 Availability during Hemodilution

1984

An insufficient and heterogeneously distributed nutritive blood flow leads to an inadequate and nonuniform supply of O2 and substrates in many solid tumors (Vaupel, 1977, 1979, 1982). This deterioration of the supply conditions, which already occurs in very early growth stages and which is superimposed by a deterioration of diffusive transport during advanced growth stages, is paralleled by a decrease in the therapeutic efficacy of various cancer treatment modalities such as irradiation and chemotherapy with antiproliferative drugs. In the case of anticancer drugs, the efficiency may be reduced by affecting both pharmacokinetics and pharmacodynamics. In the latter case this is due to the de…

Advances in understanding mineral dust and boundary layer processes over the Sahara from Fennec aircraft observations

2015

Abstract. The Fennec climate programme aims to improve understanding of the Saharan climate system through a synergy of observations and modelling. We present a description of the Fennec airborne observations during 2011 and 2012 over the remote Sahara (Mauritania and Mali) and the advances in the understanding of mineral dust and boundary layer processes they have provided. Aircraft instrumentation aboard the UK FAAM BAe146 and French SAFIRE (Service des Avions Français Instrumentés pour la Recherche en Environnement) Falcon 20 is described, with specific focus on instrumentation specially developed for and relevant to Saharan meteorology and dust. Flight locations, aims and associated met…

Data flow of spectral UV measurements at Sodankylä and Jokioinen

2016

Abstract. The data flow involved in a long-term continuous solar spectral UV irradiance monitoring program is investigated and structured to provide an overall view on the multiphase process from data acquisition to the final products. The program employing Brewer spectrophotometers as measuring instruments is maintained by the Finnish Meteorological Institute (FMI) ever since the 1990s at two sites in Finland: Sodankylä (67° N) and Jokioinen (61° N). It is built upon rigorous operation routines, processing procedures, and tools for quality control (QC) and quality analysis (QA) under continuous development and evaluation. Three distinct levels of data emerge, each after certain phase in th…

The primary volcanic aerosol emission from Mt Etna: Size-resolved particles with SO2 and role in plume reactive halogen chemistry

2018

International audience; Volcanoes are an important source of aerosols to the troposphere. Within minutes after emission, volcanic plume aerosol catalyses conversion of co-emitted HBr, HCl into highly reactive halogens (e.g. BrO, OClO) through chemical cycles that cause substantial ozone depletion in the dispersing downwind plume. This study quantifies the sub-to-supramicron primary volcanic aerosol emission (0.2-5 μm diameter) and its role in this process. An in-situ ground-based study at Mt Etna (Italy) during passive degassing co-deployed an optical particle counter and Multi-Gas SO2 sensors at high time resolution (0.1 Hz) enabling to characterize the aerosol number, size-distribution and…

An integrated energy simulation and life cycle assessment to measure the operational and embodied energy of a Mediterranean net zero energy building

2022

Abstract Net Zero Energy Buildings can play a key role in reducing the energy use, greenhouse gas emissions and the use of natural resources associated to the built environment. It is widely recognized that in low energy building a shift in relevance may occur from the operational to the embodied impacts. However, building performance assessment focus mostly on the operational phase and on primary energy consumption. This study aims to assess the energy and environmental impacts of a residential net zero energy building expanding the analysis to the full life cycle and to a wide range of environmental impact categories. The methodological approach integrates a non-steady state building ener…

Donor CD4 T cells convert mixed to full donor T-cell chimerism and replenish the CD52-positive T-cell pool after alemtuzumab-based T-cell-depleted al…

2009

Donor lymphocyte infusions (DLI) are used to resolve mixed T-cell chimerism (TCC) after allo-SCT despite a substantial risk of GVHD. We analyzed the impact of prophylactic CD8-depleted (CD8(depl)) DLI in 20 recipients of anti-CD52 alemtuzumab in vivo T-cell-depleted allografts with declining donor TCC after day +60. A total of 13 patients received CD8(depl) DLI and 7 patients did not. All but one of the DLI patients converted to complete donor T-cell chimeras, whereas only one non-DLI patient converted spontaneously. DLI induced transient acute GVHD in five and extensive chronic GVHD in two patients. These data suggest the use of CD8(depl) DLI as an effective treatment for mixed TCC, partic…
