Rat liver microsomes which were induced either with ethanol or PB were incubated with NDMA or NMA. Formaldehyde generation and nitrite formation were measured as metabolic parameters for oxidative bioactivation and denitrosation, respectively. The influence of antiserum PB3a1 and PB22 containing antibodies against the corresponding cytochrome P-450 species on both metabolic functions was investigated. The results showed that the influence on formaldehyde production and denitrosation varied independently in that both parameters were either not affected, or influenced in an opposite way, or inhibited to a different degree. Abstract A new method is presented for the determination of DMA in water as its 9-fluorenylmethyl chloroformate (FMOC) derivative using solid-phase microextraction (SPME) and liquid chromatography. The method is based on the employment of SPME fibres coated with carbowax-templated resin (CW-TR) for analyte extraction and derivatization. The fibres were successively immersed in the samples, in a solution of o -phthalaldialdehyde and N -acethyl- l -cysteine (OPA–NAC) and finally, in a solution of FMOC. Several new 4-(3,3-dimethyltriazeno)-5-benzamidopyrazole derivatives were prepared by reacting 4-diazo-5-benzamidopyrazole derivatives with dimethylamine. The compounds were tested at 10 microM for their vitro antileukemic activity against K562 (Human chronic myelogenous leukemia) and Raji (human Burkitt limphoma ) cell lines. Dacarbazine and methotrexate were used for comparative purpose. The 3-methyl-4-(3,3-dimethyltriazeno)-5-(substituted benzamido)pyrazoles, bearing the pyrazole nucleus free at 1 position, resulted more active than the 1-(substituted phenyl)-3-methyl-4-(3,3-dimethyltriazeno)-5-benzamidopyrazoles. Synthesis and inhibitory activity of dimethylamino-chalcone derivatives on the induction of nitric oxide synthase.

A series of nine dimethylamino-chalcone derivatives (1,3-diaryl-propenones) was synthesized and screened as potential inhibitors of NO and PGE(2) production in the RAW 264.7 macrophage cell line. 4-Dimethylamino-2',5'-dimethoxychalcone (6) was found to be the most potent and dual inhibitor (IC(50s) in the submicromolar range) of NO and PGE(2) production. 2',6'-Dimethoxylation appeared to be an effective requirement for selective and potent inhibition of nitric oxide synthase induction as it was confirmed by Western blot analysis. Chalcone (6) at 25 mg kg(-1) by oral route, inhibited significantly the formation of oedema in the carrageenan-induced model of inflammation in mice.
Chemical recycling of polyenaminones by transamination reaction via amino–enaminone polymerisation/depolymerisation

Abstract Novel oligo- and polyenaminones with Mw ∼ 3000 Da were prepared in excellent yields by amino–enaminone ‘click’ polymerisation of 1,3- and 1,4-bis[3-(dimethylamino)acryloyl]arenes with 1,3- and 1,4-phenylenediamine. The obtained oligomers and polymers were depolymerised quantitatively to the starting bis-enaminones and diamines by treatment with excess dimethylamine in ethanol at 50 °C. This simple polymerisation/depolymerisation method allows for a closed-loop recycling of polyenaminone-based materials. UV light absorbance at wavelengths below 470 nm indicates promising optical properties of these polymers.

The reliability of SPME combined with a chemical reaction for the analysis of short-chain aliphatic amines by liquid chromatography has been investigated. Different options to couple SPME and derivatization have been tested and compared: (i) derivatization of the analytes in solution followed by the extraction of the derivatives, (ii) extraction of the analytes and subsequent derivatization by immersing the SPME fibre onto a solution of the reagent, and (iii) extraction/derivatization of the analytes using fibres previously coated with the reagent. Synthesis and antiproliferative activity of triazenoindazoles and triazenopyrazoles: a comparative study.

Several triazenoindazoles and triazenopyrazoles were prepared transforming the appropriate aminoindazoles and aminopyrazoles in the corresponding diazonium salts which were reacted with dimethylamine, diethylamine and pyrrolidine. All the triazenes were tested for their antiproliferative activity against K562, HL60, L1210 and MCF7 cell lines. The biological data showed that the benzocondensation plays a positive role on the antiproliferative activity. The (1)H-NMR spectra showed that the rotational barrier around the N(2)-N(3) bond in the triazene group can be influenced both by the position of this group in the indazole nucleus and by the substitution pattern in the benzene moiety.
Preparation of 2-amino-5-methyl-7H-1,3,4-thiadiazolo[3,2-α]pyrimidin-7-ones

2-Amino substituted 7H-1,3,4-thiadiazolo[3,2-α]pyrimidin-7-ones 11a-e were prepared by the reaction of 2-bromo-5-amino-1,3,4-thiadiazole (1b) and diketene (8), subsequent cyclocondensation (9b 3b) and displacement of the bromo substituents by the reaction with primary or secondary amines (3b 11a-e). The hydrogen atom 6-H in the heterobicycle 3b is replaced by a Cl or Br atom in the transformation of 3b 14a,b. The 2-bromo-6-chloro compound 14a reacts chemoselectively in the 2-position with dimethylamine (14a 15). The structure elucidations are based on one- and two-dimensional NMR techniques including a heteronuclear NOE measurement.
Enthalpies of transfer of pentanol from water to sodium dodecylsulfate-dodecyl-dimethylamine oxide-water mixtures

At a given surfactant-surfactant ratio, the enthalpies of transfer ΔH (W→W+S) of pentanol 0.03m from water to sodium dodecylsulfate (NaDS)-dodecyldimethylamine oxide-water mixtures as functions of the surfactants mixture concentration (m t) were determined. ForX NaDS=0.9, ΔH (W→W+S) increases monotonically withm t such as observed for pure surfactants. ForX NaDS=0.12 and 0.3, ΔH (W→W+S) increases withm t up to 0.12m beyond which it decreases withm t. AtX NaDS=0.6, two monotonic curves can be distinguished in the ΔH (W→W+S)vs. m t trend.
