MAST-RT0 solution of 3D Navier Stokes equations in very irregular domains. Preliminary results in the laminar case

A new numerical methodology to solve the 3D Navier-Stokes equations for incompressible fluids within complex boundaries and unstructured body-fitted tetrahedral mesh is presented and validated with three literature and one real-case tests. We apply a fractional time step procedure where a predictor and a corrector problem are sequentially solved. The predictor step is solved applying the MAST (Marching in Space and Time) procedure, which explicitly handles the non-linear terms in the momentum equations, allowing numerical stability for Courant number greater than one. Abstract C75: Overcoming KRAS/LKB1 mutant NSCLC resistance to BET bromodomain inhibitors with gemcitabine or Mcl-1 inhibition

Abstract The purpose of our study was to define a method and mechanism for overcoming the resistance of clinically relevant KRAS-mutant/LKB1-deficient NSCLC cells to the BET-bromodomain inhibitor JQ1. LKB1 (Serine/threonine kinase 11) is mutated with loss of function in conjunction with mutated KRAS in 7-10% of NSCLC. Importantly, KRAS-mutant/LKB1-deficiency is associated with tumor aggressiveness and poor survival in human patients as well as in genetically engineered mouse models. Indeed, although the BET bromodomain inhibitor JQ1 dramatically reduces tumor volume in KRAS mutant mice, it has little effect in KRAS-mutant/LKB1-deficient mice. Down-regulation of Endogenous Amyloid Precursor Protein Processing due to Cellular Aging

Processing of amyloid precursor protein (APP) is a well acknowledged central pathogenic mechanism in Alzheimer disease. However, influences of age-associated cellular alterations on the biochemistry of APP processing have not been studied in molecular detail so far. Here, we report that processing of endogenous APP is down-regulated during the aging of normal human fibroblasts (IMR-90). The generation of intracellular APP cleavage products C99, C83, and AICD gradually declines with increasing life span and is accompanied by a reduced secretion of soluble APP (sAPP) and sAPPalpha. The best-characterized membrane trafficking organelles include the endoplasmic reticulum (ER), Golgi apparatus, early and recycling endosomes, multivesicular body, or late endosome, lysosome/vacuole, and plasma membrane. Although historically plants have given rise to cell biology, our understanding of membrane trafficking has mainly been shaped by the much more studied mammalian and yeast models. This paper shows an investigation on optimal damping constant performed in the frequency domain. The optimal damping constant is meant as that value that minimizes the acceleration of all connected bodies characterizing a two degree of freedom system sketching a quarter car. The connected bodies are sprung and unsprung mass respectively for quarter of chassis and tire, this last keeps the contact with the ground and it is connected with the sprung mass through a shock absorber characterized by spring and fluid damper.
