Comparative study assessing effects of sonic hedgehog and VEGF in a human co-culture model for bone vascularisation strategies.

2011

The morphogen sonic hedgehog (Shh) seems to mediate adult repair processes in bone regeneration and vascularisation. In this study we investigated the effects of Shh on co-cultures consisting of human primary osteoblasts and outgrowth endothelial cells in terms of angiogenic activation and vessel maturation in comparison to the treatment with the commonly used proangiogenic factor, VEGF. Both, stimulation with VEGF or Shh, leads to an increase in the formation of microvessel-like structures compared to untreated controls. In contrast to VEGF, proangiogenic effects by Shh could already be observed after 24 h of treatment. Nevertheless, after 14 days the angiogenic activity of OEC was compara…

0393: Impact of miR-378* and its target desmin intermediate filament on mitochondria distribution in cardiomyocytes

2014

Background MiR-378 and miR-378* microRNAs are derived from an intron of the PGC-1β gene, a regulator of mitochondrial biogenesis. Their expression is either repressed or increased during heart failure depending on the model. Through proteomics approaches, we previously identified new targets of these miRs in H9c2 fetal cardiomyoblasts, among which lactate dehydrogenase for miR-378 and key cytoskeletal proteins for miR-378*. Aims To better assess its role in energy metabolism and differentiation; we overexpressed miR-378 and miR-378* in primary neonate rat cardiomyocytes (NRC) that are more differentiated and less proliferative than H9c2 cardiomyoblasts. Results We identified desmin as a new…

Tenascin gene expression in rat liver and in rat liver cells. In vivo and in vitro studies.

1991

Tenascin is a major glycoprotein constituent of the extracellular matrix with a strong affinity to fibronectin; its distribution is believed to be temporarily and spatially limited. Tenascin gene expression is increased during wound healing processes. As repair mechanisms in chronic liver diseases resemble wound healing we studied tenascin gene expression in rat liver and in isolated rat liver cells. In normal rat liver a tenascin specific antiserum stains sinusoidal cells with fiber-like prolongations, which at the same time are desmin-positive (ITO-cells). In the CCl4-acutely-damaged liver a strong tenascin staining is detected in cells located among the mononuclear cells of the inflammat…

Gender differences in skeletal muscle fibre damage after eccentrically biased downhill running in rats

1999

Specific antibodies against structural proteins of muscle fibres (actin, desmin, dystrophin) and extracellular matrix (fibronectin) were used to study the effect of eccentrically biased downhill running exercise (13,5 degrees, 17 m min(-1), 130 min) on the magnitude and properties of myofibre injury in the quadriceps femoris muscle of male and female rats. Muscle beta-glucuronidase activity, a quantitative indicator of muscle damage, showed clearly smaller increase in female than in male rats during the 4-day period following exercise. A similar course of histopathological changes was observed in both sexes, although females showed slower and less marked changes than males. In males, discon…

Potential role of the neuropeptide CGRP in the induction of differentiation of rat hepatic portal vein wall.

2005

The media of the rat hepatic portal vein is composed of an internal circular muscular layer (CL) and an external longitudinal muscular layer (LL). These two perpendicular layers differentiate progressively from mesenchymal cells within the first month after birth. In this paper, we studied the development of calcitonin gene-related peptide (CGRP) innervation during post-natal differentiation of the vessel. We show that CGRP innervation is already present around the vessel at birth in the future adventitia but far from the lumen of the vessel. Progressively, CGRP immunoreactive fibers reached first LL then CL. CL by itself become only innervated at day 14 after birth. This corresponds to the…

Elevated intraocular pressure induces neuron-specific β-III-tubulin expression in non-neuronal vascular cells.

2019

Purpose Pathological alterations within optic nerve axons and progressive loss of the parental retinal ganglion cell (RGC) bodies are characteristics of glaucomatous neuropathy. Abnormally elevated intraocular pressure (IOP) is thought to be the major risk factor for most forms of glaucomatous changes, while lowering of the IOP is the mainstream of treatment. However, the pathophysiological mechanisms involved in neurodegenerative changes are poorly understood. It remains still a matter of debate whether elevated IOP harms the neurons directly or indirectly through alterations in the retinal vascularization. Methods We analysed morphological and molecular changes within the retina exposed t…
