Osteoprotegerin expression in liver is induced by IL-13 through TGF-β

2020

1.AbstractBackgroundsOsteoprotegerin (OPG) is a profibrotic mediator produced by myofibroblasts under influence of transforming growth factor β (TGFβ). Its expression in experimental models of liver fibrosis correlates well with disease severity and treatment responses. The regulation of OPG in liver tissue is largely unknown and we therefore set out to elucidate which growth factors/interleukins associated with fibrosis induce OPG and through which pathways.MethodsPrecision-cut liver slices of wild type and STAT6-deficient mice and 3T3 fibroblasts were used to investigate the effects of TGFβ, interleukin (IL) 13 (IL13), IL1β, and platelet-derived growth factor BB (PDGF-BB) on expression of…

Muscleblind, BSF and TBPH are mislocalized in the muscle sarcomere of a Drosophila myotonic dystrophy model

2012

SummaryMyotonic dystrophy type 1 (DM1) is a genetic disease caused by the pathological expansion of a CTG trinucleotide repeat in the 3' UTR of the DMPK gene. In the DMPK transcripts, the CUG expansions sequester RNA-binding proteins into nuclear foci, including transcription factors and alternative splicing regulators such as MBNL1. MBNL1 sequestration has been associated with key features of DM1. However, the basis behind a number of molecular and histological alterations in DM1 remain unclear. To help identify new pathogenic components of the disease, we carried out a genetic screen using a Drosophila model of DM1 that expresses 480 interrupted CTG repeats, i(CTG)480, and a collection of…

Type V collagen counteracts osteo-differentiation of human mesenchymal stem cells

2014

In search of novel gene signatures for osteo-differentiation of mesenchymal stem cells (MSCs), we submitted cDNA preparations from undifferentiated and differentiating MSCs to differential display- and semiquantitative-PCR and found down-regulation of COL5A1 in osteo-induced cultures at days 21 and 28, when the mineralized matrix accumulates. We also cultured osteo-differentiating MSCs onto type V collagen substrates and found a decrease in the accumulation of extracellular calcium compared to those grown in uncoated flasks. To our knowledge, this is first evidence that type V collagen might represent a stromal component that impairs osteogenesis.

The Epidemiology of Hip and Knee Primary and Revision Arthroplasties during the COVID-19 Pandemic

2021

Background: The purpose of this study was to provide a comprehensive assessment of the impact of the COVID-19 pandemic on the epidemiology of primary and revision arthroplasties of the hip and knee joint. Methods: This study compared the data on knee and hip arthroplasty procedures from 2 hospitals (primary and revision) conducted in two periods: the period of the COVID-19 pandemic in Poland (from 4 March 2020 to 15 October 2020) and the corresponding period prior to the pandemic (from 4 March 2019 to 15 October 2019). We compared the epidemiological data, demographic data, and hospital stay duration data from these two periods. Results: Our analysis demonstrated that the total number of hi…

Realne i cudowne elementy w narracji o dziejach pierwszej krucjaty (na przykładzie Gesta Francorum)

2018

Pierwsza krucjata była z pewnością jednym z przełomowych wydarzeń europejskiego średniowiecza, a opowieści o niej ukształtowały kolejne pokolenia mieszkańców starego kontynentu.

High-Fat Diet Induces Pre-Diabetes and Distinct Sex-Specific Metabolic Alterations in Negr1-Deficient Mice

2021

In the large GWAS studies, NEGR1 gene has been one of the most significant gene loci for body mass phenotype. The purpose of the current study was to clarify the role of NEGR1 in the maintenance of systemic metabolism, including glucose homeostasis, by using both male and female Negr1−/− mice receiving a standard or high fat diet (HFD). We found that 6 weeks of HFD leads to higher levels of blood glucose in Negr1−/− mice. In the glucose tolerance test, HFD induced phenotype difference only in male mice

Dynamic shakedown design of structures under repeated seismic loads

2013

The paper is devoted to the formulation of an optimal design (minimum volume) problem of elastic perfectly plastic structures subjected to suitable combinations of static (fixed) and dynamic (seismic) loads. The structure is constrained to simultaneously respect two different safety criteria; actually, it must exhibit an elastic shakedown behaviour for the combination of loads characterizing the serviceability conditions and it must prevent the instantaneous collapse for the highest expected load condition (combination of loads characterized by the presence of fixed loads and maximum expected intensity of seismic action). The shakedown limit behaviour for the optimal structure will be impos…
