Retrotransposon silencing and telomere integrity in somatic cells of Drosophila depends on the cytosine-5 methyltransferase DNMT2

Here we show that the cytosine-5 methyltransferase DNMT2 controls retrotransposon silencing in Drosophila somatic cells. In Drosophila, significant DNMT2-dependent DNA methylation occurs during early embryogenesis. Suppression of white gene silencing by Mt2 (Dnmt2) null mutations in variegated P[w(+)] element insertions identified functional targets of DNMT2. The enzyme controls DNA methylation at retrotransposons in early embryos and initiates histone H4K20 trimethylation catalyzed by the SUV4-20 methyltransferase. Intraspecific variation in mating behaviour does not cause sexual isolation between Drosophila virilis strains

There is increasing evidence that mate recognition systems vary within species. We studied the courtship rituals of five Drosophila virilis strains to investigate this intraspecific variation. Males that actively and persistently courted the female without paying much attention to the female's behaviour were the most successful at copulating. Intraspecific variation in mating rituals was high, but not as high as at the species level and it was not high enough to cause sexual isolation between the strains. Immunological detection of phenylalanine hydroxylase protein in Drosophila melanogaster.

A monoclonal antibody raised against monkey liver phenylalanine hydroxylase (PAH) has been used to detect this protein in Drosophila melanogaster. A cross-reacting material (CRM) band of apparent molecular mass 50-52 kDa, equivalent to that deduced for the Drosophila melanogaster PAH protein based on the pah gene cDNA sequence, has been detected. This CRM was analysed throughout development and showed an equivalent pattern to that reported for PAH activity in this insect, with maxima at pupariation and at pharate adult formation. Comparative evolution of P–M system and infection by the sigma virus in French and Spanish populations ofDrosophila melanogaster

SummaryIn 1983, an extensive survey of populations ofD. melanogasterwas started in a southern French region (Languedoc) in two non-Mendelian systems: the P–M system of transposable elements and the hereditary Rhabdovirus sigma. Unexpectedly fast-evolving phenomena were observed and interesting correlations were noted, giving similar geographical pattern to the region in both systems. For these reasons, the analysis was continued and extended towards the north (Rhône Valley) and the south (Spain). Effects of cold acclimation and dsRNA injections on Gs1l gene splicing in Drosophila montana

Abstract Alternative splicing, in which one gene produce multiple transcripts, may influence how adaptive genes respond to specific environments. A newly produced transcriptome of Drosophila montana shows the Gs1-like (Gs1l) gene to express multiple splice variants and to be down regulated in cold acclimated flies with increased cold tolerance. Gs1l’s effect on cold tolerance was further tested by injecting cold acclimated and non-acclimated flies from two distantly located northern and southern fly populations with double stranded RNA (dsRNA) targeting Gs1l. The banding pattern of polytene chromosomes of Drosophila guanche compared with that of D. subobscura.

A detailed map of the salivary gland chromosomes of Drosophila guanche is presented and compared to the standard gene arrangements of D. subobscura. Generally, the polytene chromosomc banding patterns of the two species show a high degrce of homology. Only Segment I of the sex chromosome (Chromosome A) shows marked differences. The banding pattern proposed for this segment in D. guanche could have originated from a cluster of overlapping inversions including A1 arrangement.
