Tetraspanins (Tspans) are a family of four-span transmembrane proteins, known as plasma membrane “master organizers.” They form Tspan-enriched microdomains (TEMs or TERMs) through lateral association with one another and other membrane proteins. If multiple microdomains associate with each other, larger platforms can form. For infection, viruses interact with multiple cell surface components, including receptors, activating proteases, and signaling molecules. It appears that Tspans, such as CD151, CD82, CD81, CD63, CD9, Tspan9, and Tspan7, coordinate these associations by concentrating the interacting partners into Tspan platforms. Many pathogens, including viruses, bacteria, as well as bacterial toxins, enter their target cells by endocytosis leading to accumulation of pathogenic and cellular proteins in endosomes. Here, we present detailed experimental instructions on isolation of endosomes after virus infection and their subsequent biomolecular characterization. The isolation of endosomes is based on discontinuous sucrose gradient centrifugation, where different endosomal compartments accumulate at a specific sucrose interface. The advances of short interfering RNA (siRNA)-mediated therapy provide a powerful option for the treatment of many diseases, including cancer, by silencing the expression of targeted genes involved in the progression of the pathology. On this regard, a new pH-responsive polycation derived from inulin, Inulin-g-imidazole-g-diethylenetriamine (INU-IMI-DETA), was designed and employed to produce INU-IMI-DETA/siRNA "Inulin COmplex Nanoaggregates" (ICONs). Kirsi Pakkanen haki väitöstutkimuksessaan monitieteellisen lähestymistavan avulla uusia näkökulmia virustutkimukseen. Tutkimus tuo uutta tietoa lipidikalvojen merkityksestä viruksen ja viruksen isäntäsolun elämässä. Tutkimuksessa selvisi, että koiran parvovirus, jota tutkimuksessa käytettiin mallina parvoviruksille ja yleisemmin vaipattomille viruksille, tarvitsee tietynlaisia isäntäsolun lipidikalvojen ominaisuuksia voidakseen vapautua solunsisäisten endosomirakkuloiden sisältä.
