Oxymetazoline modulates proinflammatory cytokines and the T‐cell stimulatory capacity of dendritic cells

2007

The nasal decongestant oxymetazoline (OMZ) is frequently used in the topical treatment of rhinitis/sinusitis. As proinflammatory cytokines play a critical role in the development and maintenance of local inflammation, the aim of this study was to investigate the influence of OMZ on immune cells in order to diminish the mucosal infiltration of the nose. Peripheral blood mononuclear cells (PBMC) from buffy coats of healthy volunteers were isolated and stimulated in the presence or absence of OMZ. In addition, monocyte-derived dendritic cells (DC) were generated and different concentrations of OMZ were added. DC phenotype and their T-cell stimulatory properties were analysed. The vasoactive su…

Hepatocellular cancer cell lines, Hep-3B and Hep-G2 display the pleiotropic response to resveratrol and berberine

2022

Purpose Human carcinoma cells with different p53 status exposed to a combination of bioactive substances, resveratrol and berberine, revealed different responses in cell viability via p53-dependant apoptosis pathway activation. Materials and methods Using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay, we investigated various and opposing effects in hepatocellular carcinoma cells, Hep-G2 and Hep-3B with different p53-status. Results Cells decreased in viability after treatment with dose-dependent concentrations of resveratrol and berberine. Hep-3B p53 mutants were more sensitive in comparison to the p53 wild type Hep-G2 cell line. A syne…

Influence of aryl hydrocarbon- (Ah) receptor and genotoxins on DNA repair gene expression and cell survival of mouse hepatoma cells

2009

The aryl hydrocarbon receptor (AhR) mediates toxicity of a variety of environmental pollutants such as polycyclic aromatic hydrocarbons (PAHs) and dioxins. However, the underlying mechanisms and genetic programmes regulated by AhR to cause adverse effects but also to counteract poisoning are still poorly understood. Here we analysed the effects of two AhR ligands, benzo[a]pyrene (B[a]P), a DNA damaging tumour initiator and promotor and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), a pure tumour promoter, on cell survival and on nucleotide excision repair (NER) gene expression. NER deals with so called "bulky" DNA adducts including those generated by enzymatically activated B[a]P. Therefore, t…

Targeting HSP90 with the small molecule inhibitor AUY922 (luminespib) as a treatment strategy against hepatocellular carcinoma

2018

Hepatocellular carcinoma (HCC) is a highly malignant tumor that responds very poorly to existing therapies, most probably due to its extraordinary inter- and intra-tumor molecular heterogeneity. The modest therapeutic response to molecular targeted agents underlines the need for new therapeutic approaches for HCC. In our study, we took advantage of well-characterized human HCC cell lines, differing in transcriptomic subtypes, DNA mutation and amplification alterations, reflecting the heterogeneity of primary HCCs, to provide a preclinical evaluation of the specific heat shock protein 90 (HSP90) inhibitor AUY922 (luminespib). Indeed, HSP90 is highly expressed in different tumor types, but it…

Methylprednisolone-induced hepatotoxicity in a 16-year-old girl with multiple sclerosis.

2018

Multiple sclerosis (MS) is a chronic inflammatory disease with demyelination of the central nervous system. High-dosage corticosteroids are the first-line therapy in the acute relapsing of MS. We report a case of severe high-dose methylprednisolone-induced acute hepatitis in a patient with a new diagnosis of MS. A 16-year-old girl was admitted for urticaria, angioedema, nausea and vomiting a month later she had been diagnosed with MS and treated with high-dosage methylprednisolone. Laboratory investigations showed hepatic insufficiency with grossly elevated liver enzymes. A liver biopsy showed focal centrilobular hepatocyte necrosis with interface hepatitis. Methylprednisolone-induced hepat…

Itsepalvelukassat asiakasnäkökulmasta: Vaikutus asiakaskokemuksen muodostumisessa

2014

Tutkielma selvittää aiempaan tutkimukseen perustuen itsepalvelukassojen ja asiakaskokemuksen suhdetta. Tavoitteena on hahmottaa, kuinka itsepalvelukassat vaikuttavat asiakaskokemuksen muodostumisessa. Lisäksi selvitetään edellytykset itsepalvelukassojen hyvän asiakaskokemuksen aikaansaamiseksi. Tutkimustarve perustuu nousevaan kiinnostukseen itsepalveluteknologioita kohtaan, kuten myös asiakaskokemuksen merkityksen kasvulle kaupanalan kilpailun kiristyessä ja asiakkaiden vaatimusten kasvaessa. Lisäksi tutkimus on aiemmin painottunut lähinnä itsepalvelukassojen teknisiin ja taloudellisiin edellytyksiin, sekä käytettävyyden tarkasteluun. Tutkielma yhdistelee näkökulman uutuuden vuoksi laajast…

Achieving agreement on service needs in child protection : Comparing children’s, mothers’ and practitioners’ views over time and between approaches

2022

Purpose While studies on service users’ participation and their perceptions on the quality of services exist, agreement between family members’ and practitioners’ assessments of the family’s situation has received less interest. The purpose of this paper is to investigate agreement and its effect on outcomes by comparing the viewpoints of three groups of informants (children, mothers and practitioners) in the context of statutory child protection in two study groups – one applying a systemic approach (SPM) and a service-as-usual control group (SAU). Design/methodology/approach A quasi-experimental repeated-measures study design was applied. Outcome data comprised 112 cases (SPM cases n = 5…

Publisko iepirkumu nepamatotas sadalīšanas regulējuma problemātika Latvijā un Eiropas Savienībā

2022

Maģistra darbā “Publisko iepirkumu nepamatotas sadalīšanas regulējuma problemātika Latvijā un Eiropas Savienībā” tiek apskatītas būtiskākās ar publisko iepirkumu nepamatotas sadalīšanas tiesisko regulējumu saistītās problēmas publisko iepirkumu procesā, kā arī ieteikts iespējamais šo problēmu risinājums. Par tēmas aktualitāti liecina tas, ka neatbilstošas iepirkuma procedūras izvēle vai publisko iepirkumu regulējuma nepiemērošana vispār ir viens no būtiskākajiem publisko iepirkumu pārkāpumiem, un pie tā bieži noved kļūdas iepirkuma plānošanā, tai skaitā nepamatota iepirkuma sadalīšana un nepareizas paredzamās līgumcenas noteikšana. Maģistra darba mērķa sasniegšanai veikta iepirkumu nepamato…

Party autonomy regarding jurisdiction under the property regimes regulations

2021

The main accomplishment of the Property Regimes Regulations lies in their bringing more coherence into the cross-border family law adjudication. In the field of international jurisdiction, they strive to align the competence in couples? patrimony disputes to that in succession and in separation proceedings, or else to align the competence of the courts to the applicable law. These tendencies are clearly visible in the Regulations? provisions on choice of court agreements. Namely, the Regulations allow for such agreements, but severely limit parties? choice and the possible effects of these clauses.
