Synthesis and mutagenicity of the diastereomeric fjord-region 11,12-dihydrodiol 13,14-epoxides of dibenzo[a,l]pyrene.

1994

Extensive tumorigenicity studies in rodents revealed that dibenzo[a,l]pyrene (DB[a,l]P) is the most potent carcinogen among all polycyclic aromatic hydrocarbons (PAHs) tested so far. The structure of the genotoxic metabolite(s) responsible for this exceptional carcinogenicity is unknown. The fjord-region syn- and anti-DB[a,l]P-11,12-dihydrodiol 13,14-epoxides (syn- and anti-DB[a,l]PDE) were synthesized to clarify their role as possible ultimate mutagenic and carcinogenic metabolites of DB[a,l]P.9-Formyl-11,12-dimethoxybenzo[g] chrysene was prepared from 9-phenanthrylacetic acid by a photochemical route. After reaction of the aldehyde with trimethylsulfonium iodide to generate an oxiranyl si…

Loss of circadian clock gene expression is associated with tumor progression in breast cancer

2014

Several studies suggest a link between circadian rhythm disturbances and tumorigenesis. However, the association between circadian clock genes and prognosis in breast cancer has not been systematically studied. Therefore, we examined the expression of 17 clock components in tumors from 766 node-negative breast cancer patients that were untreated in both neoadjuvant and adjuvant settings. In addition, their association with metastasis-free survival (MFS) and correlation to clinicopathological parameters were investigated. Aiming to estimate functionality of the clockwork, we studied clock gene expression relationships by correlation analysis. Higher expression of several clock genes (e.g., C…

Influence of pH on the benzodiazepine-human serum albumin complex. Circular dichroism studies.

1974

The influence of pH on the binding of benzodiazepine derivatives to HSA was studied by circular dichroism measurements and by gel filtration. The binding of nearly all benzodiazepines is increased by rising the pH from 6.60 to 8.20. For flurazepam, clonazepam, and nitrazepam this increase in binding is due to an increase of the affinities, while for the other substances the affinity remains constant and the number of binding sites is increased from one to two. The changes in binding of the benzodiazepines by rising the pH are explained by a cationic amino acid residue near or at the benzodiazepine binding site of the HSA molecule. This second binding site is not detectable by circular dichr…

Studies on Adduct Formation of (+)-Anti-Benzo[a]pyrene 7,8-Dihydrodiol 9,10-Epoxide with the oligonucleotides 5′-d(CCTATCGTTATCC) and 5′-d(CCTATm5CGT…

1999

Abstract Adduct formation of (+)-anti-benzo[a]pyrene 7,8-dihydrodiol 9,10-epoxide [BPDE] and 5′-d(CCTATCGTTATCC) or 5′-d(CCTATm5CGTTATCC) (G = binding target) has been studied. The extent of trans-BPDE-N2-dG adduct formation was higher in the oligonucleotide with 5′-d(m5CG) sequence context in both single- and double stranded form compared to the non-methylated analogue. The stimulating effect of m5dC on adduct formation has previously been demonstrated in other experimental systems. The increase in yield could possibly be rationalized in terms of prestacking of the pyrenyl ring with the nucleobases prior to the nucleophilic addition. In the present study, both UV absorption and induced cir…

Interferon alfa–induced sarcoidosis resolving without drug withdrawal

2016

Sarcoidosis is an uncommon systemic granulomatous disease of unknown origin affecting lung, skin, liver, and other tissues. Noncaseating granulomas in the involved organs are the hallmark of this disease. An exaggerated immune response to an unknown antigenic stimulus could play a role in sarcoidosis development. Lung is one of the most frequently involved organs.1 Manifestations range from alveolitis to granulomatous infiltration of alveoli, bronchi, and blood vessels. The end stage of lung sarcoidosis is development of interstitial fibrosis with “honeycombing” of lung parenchyma. Interferon alfa in association with ribavirin is the treatment of choice for hepatitis C. Early treatment of a…

Use of Ultrasound in Reconditioning by Welding of Tools Used in the Process of Regenerating Rubber

2018

Addressing the problem of reconditioning large parts is of particular importance, due to their value and to the fact that the technologies for their reconditioning are very complex. The tools used to refine regenerated rubber which measure 660 mm in diameter and 2130 mm in length suffer from a rather fast dimensional wear. Within this research, the authors looked for a welding reconditioning procedure that would allow a very good adhesion between the deposited material layer and the base material. In this regard, the MAG (Metal Active Gas) welding process was used, but the ultrasonic activation of the welding process was also considered. Thus, the wire used for welding was activated conside…
