Marked Neurospora crassa strains for competition experiments and Bayesian methods for fitness estimates

2019

AbstractThe filamentous fungusNeurospora crassa, a model microbial eukaryote, has a life cycle with many features that make it suitable for studying experimental evolution. However, it has lacked a general tool for estimating relative fitness of different strains in competition experiments. To remedy this need, we constructedN. crassastrains that contain a modifiedcsr-1locus and developed an assay for detecting the proportion of the marked strain using a post PCR high resolution melting assay. DNA extraction from spore samples can be performed on 96-well plates, followed by a PCR step, which allows many samples to be processed with ease. Furthermore, we suggest a Bayesian approach for estim…

Temporal and spatial patterns of trawl fishing activities in the Adriatic Sea (Central Mediterranean Sea, GSA17)

2020

Abstract Trawl fishing activities have occurred for centuries on large spatial scale in the entire Mediterranean Sea, and today they are considered as one of the main and widespread causes of anthropogenic disturbance and habitat alteration in the marine environment. In order to delineate when, where and how marine ecosystems have been perturbed and to implement ecosystem-based management strategies, the identification and investigation of the spatial and temporal distribution of fishing effort and the fleet dynamics play a key role. In this context, Geospatial Technologies such as the Automatic Identification System (AIS) could represent a useful tool. The aim of the present work is to rec…

High-resolution numerical modelling of the barotropic tides in the Gulf of Gabes, eastern Mediterranean Sea (Tunisia)

2017

International audience; A high-resolution 2D barotropic tidal model was developed for the Gulf of Gabes and used to characterise hydrodynamic processes and tidal dynamics. The model is based on the Regional Ocean Modelling System. It is forced at the open boundaries by the semidiurnal M2 and S2 astronomical components while meteorological forcing has been neglected. The model results show good agreement with observations confirming that it reproduces the gulf's main tidal characteristics reasonably well. In fact, the simulated semidiurnal tidal components M2 and S2 generate important sea level variations and coastal currents. Tidal propagation is directed to the gulf's western sector while …

Satellite survey of seasonal trophic status and occasional anoxic 'malaigue' crises in the Thau lagoon using MERIS images

2011

International audience; The Thau lagoon, located in southern France, suffers episodically from anoxic crises locally known as 'malaigue'. Such crises mostly occur under warm conditions, low winds leading to a strong eutrophication of the lagoon. The development of a sulphur bacterium sometimes gives locally to the waters a 'milky turquoise' appearance and leads to shellfish mortality. One of the indicators of the eutrophication status of the lagoon can be surveyed by the chlorophyll product provided by remote sensing images such as Medium Resolution Imaging Spectrometer (MERIS). In this paper we compare chl2 (or algal2) estimations provided by MERIS level 2 products and the ground measureme…

Testing macroecological abundance patterns:the relationship between local abundance and range size, range position and climatic suitability among Eur…

2020

Aim A fundamental question in macroecology centres around understanding the relationship between species' local abundance and their distribution in geographical and climatic space (i.e. the multi-dimensional climatic space or climatic niche). Here, we tested three macroecological hypotheses that link local abundance to the following range properties: (a) the abundance-range size relationship, (b) the abundance-range centre relationship and (c) the abundance-suitability relationship. Location Europe. Taxon Vascular plants. Methods Distribution range maps were extracted from the Chorological Database Halle to derive information on the range and niche sizes of 517 European vascular plant speci…

Chlorophyll a fluorescence illuminates a path connecting plant molecular biology to Earth-system science

2021

Remote sensing methods enable detection of solar-induced chlorophyll a fluorescence. However, to unleash the full potential of this signal, intensive cross-disciplinary work is required to harmonize biophysical and ecophysiological studies. For decades, the dynamic nature of chlorophyll a fluorescence (ChlaF) has provided insight into the biophysics and ecophysiology of the light reactions of photosynthesis from the subcellular to leaf scales. Recent advances in remote sensing methods enable detection of ChlaF induced by sunlight across a range of larger scales, from using instruments mounted on towers above plant canopies to Earth-orbiting satellites. This signal is referred to as solar-in…

Comprehensive analysis of photoinitiators and primary aromatic amines in food contact materials using liquid chromatography High-Resolution Mass Spec…

2018

Abstract A comprehensive strategy for the analysis of UV-ink photoinitiators and primary aromatic amines (PAAS) in food-packaging materials such as, juice tetrabricks, pouches and bags has been developed using liquid chromatography coupled to Orbitrap High-Resolution Mass Spectrometry (LC-Orbitrap-HRMS). The methodology includes both quantitative target analysis and post-run target screening analysis. The quantitative method was validated after a previous optimisation of the single-stage Orbitrap fragmentation through the Higher-Energy Collisional Dissociation (HCD) Cell. Overall, the quantitative method presented recoveries ranging from 78% to 119%, with a precision (RSD) lower than 20%, f…

Tracking Moving Objects With a Catadioptric Sensor Using Particle Filter

2011

International audience; Visual tracking in video sequences is a widely developed topic in computer vision applications. However, the emergence of panoramic vision using catadioptric sensors has created the need for new approaches in order to track an object in this type of images. Indeed the non-linear resolution and the geometric distortions due to the insertion of the mirror, make tracking in catadioptric images a very challenging task. This paper describes particle filter for tracking moving object over time using a catadioptric sensor. In this work different problems due to the specificities of the catadioptric systems such as geometry are considered. The obtained results demonstrate an…

Stabilized branch-and-price algorithms for vector packing problems

2018

Abstract This paper considers packing and cutting problems in which a packing/cutting pattern is constrained independently in two or more dimensions. Examples are restrictions with respect to weight, length, and value. We present branch-and-price algorithms to solve these vector packing problems (VPPs) exactly. The underlying column-generation procedure uses an extended master program that is stabilized by (deep) dual-optimal inequalities. While some inequalities are added to the master program right from the beginning (static version), other violated dual-optimal inequalities are added dynamically. The column-generation subproblem is a multidimensional knapsack problem, either binary, boun…

Localization microscopy of DNA in situ using Vybrant(®) DyeCycle™ Violet fluorescent probe: A new approach to study nuclear nanostructure at single m…

2016

Higher order chromatin structure is not only required to compact and spatially arrange long chromatids within a nucleus, but have also important functional roles, including control of gene expression and DNA processing. However, studies of chromatin nanostructures cannot be performed using conventional widefield and confocal microscopy because of the limited optical resolution. Various methods of superresolution microscopy have been described to overcome this difficulty, like structured illumination and single molecule localization microscopy. We report here that the standard DNA dye Vybrant(®) DyeCycle™ Violet can be used to provide single molecule localization microscopy (SMLM) images of …
