Functional significance of membrane associated proteolysis in the toxicity of Bacillus thuringiensis Cry3Aa toxin against Colorado potato beetle.

2012

Abstract Bacillus thuringiensis Cry toxins are widely used as biocontrol agents in bioinsecticides and transgenic plants. In the three domain-Cry toxins, domain II has been identified as an important determinant of their highly specific activity against insects. In this work, we assessed the role in membrane associated proteolysis and toxicity in Colorado potato beetle (CPB) of a previously reported ADAM recognition motif present in Cry3Aa toxin domain II. We used site-directed mutagenesis to modify the Bacillus thuringiensis cry3A gene in amino acid residues 344, 346, 347, 351 and 353 of the ADAM recognition motif in Cry3Aa toxin. Cry3Aa toxin mutants displayed decreased toxicity when comp…

Molecular barcoding for central-eastern European Crioceris leaf-beetles (Coleoptera: Chrysomelidae)

2011

Abstract Among Crioceris leaf-beetles, the two most widespread species (Crioceris asparagi and C. duodecimpunctata) are serious invasive plant pests, while another two (C. quatuordecimpunctata and C. quinquepunctata) are rare species restricted to steppe-like habitats in Eurasia. The aim of the research was to check the genetic distinctiveness of these four species and develop barcodes for their molecular identification using the mitochondrial Cytochrome Oxidase I (COI) gene and two nuclear markers: Elongation Factor 1-α (EF1-α) and Internal Transcribed Spacer 1 (ITS1). The identification of each species was possible and reliable with the use of COI and ITS1 markers. EF1-α was omitted in an…

First records of complete annual cycles in water rails Rallus aquaticus show evidence of itinerant breeding and a complex migration system

2020

Abstract In water rails Rallus aquaticus, northern and eastern populations are migratory while southern and western populations are sedentary. Few details are known about the annual cycle of this elusive species. We studied movements and breeding in water rails from southernmost Norway where the species occurs year-round. Colour-ringed wintering birds occurred only occasionally at the study site in summer, and vice versa. Geolocator tracks revealed that wintering birds (n = 10) migrated eastwards in spring to breed on both sides of the Baltic Sea, whereas a single breeding bird from the study site wintered in north Italy. Ambient light records of geolocator birds further indicated that all …

Unveiling gene expression regulation of the Bacillus thuringiensis Cry3Aa toxin receptor ADAM10 by the potato dietary miR171c in Colorado potato beet…

2021

Background The Colorado potato beetle (CPB) is a worldwide devastating pest of potato plants and other Solanaceae characterized by its remarkable ability to evolve resistance to insecticides. Bacillus thuringiensis (Bt) Cry3Aa toxin represents an environmentally-safe alternative for CPB control but larvae susceptibility to this toxin has been reported to vary depending on the host plant on which larvae feed. To gain more insight into how nutrition mediates Bt tolerance through effects on gene expression here we explored the post-transcriptional regulation by miRNAs of the CPB-ADAM10 gene encoding the Cry3Aa toxin functional receptor ADAM10. Results The lower CPB-ADAM10 gene expression in CP…

Pre-invasion history and demography shape the genetic variation in the insecticide resistance-related acetylcholinesterase 2 gene in the invasive Col…

2012

Abstract Background Invasive pest species offers a unique opportunity to study the effects of genetic architecture, demography and selection on patterns of genetic variability. Invasive Colorado potato beetle (Leptinotarsa decemlineata) populations have experienced a rapid range expansion and intense selection by insecticides. By comparing native and invasive beetle populations, we studied the origins of organophosphate (OP) resistance-associated mutations in the acetylcholinesterase 2 (AChE2) gene, and the role of selection and demography on its genetic variability. Results Analysis of three Mexican, two US and five European populations yielded a total of 49 haplotypes. Contrary to the exp…

Melanomas apzināšanās un riska uzvedība starp universitāšu studentiem

2016

Melanomas sastopamība turpina pieaugt visā pasaulē, it īpaši jaunu eiropeīdu pieaugušo vidū.

Citing as a sourcing practice: students’ citing self-selected online sources in their essays (Las citas como práctica del uso de las fuentes: las cit…

2020

This study examined upper secondary school students’ citations of self-selected online sources in their essays. Students (n = 140) conducted online inquiry about either effects of social media on people’s quality of life (SM) or allowance of genetic manipulation of organisms (GMO). Students, working either individually or in pairs, explored online sources with the help of a graphic organizer, after which they composed their essays. To capture the quality of citations identified in the essays, they were evaluated in terms of accuracy and richness of source features. Further, regression analysis was used to examine the effect of topic, grade level and work mode on the number and quality of ci…

Assessing the sustainability of small wastewater treatment systems: A composite indicator approach

2014

The assessment of the sustainability of wastewater treatment (WWT) systems has gained interest in recent years. However, most previous studies have focused on environmental and/or economic dimensions ignoring social aspects. Moreover, they tend to be based on sets of indicators rather than providing a holistic assessment. To overcome this limitation, this paper proposes an innovative methodology to assess the sustainability of WWT systems based on the development of a composite indicator embracing economic, environmental and social issues. Subsequently, the global sustainability of seven WWT technologies for secondary treatment in small communities is compared. The joint application of the …

Assessment of wastewater treatment alternatives for small communities: An analytic network process approach

2015

The selection of the most appropriate wastewater treatment (WWT) technology is a complex problem since many alternatives are available and many criteria are involved in the decision-making process. To deal with this challenge, the analytic network process (ANP) is applied for the first time to rank a set of seven WWT technology set-ups for secondary treatment in small communities. A major advantage of ANP is that it incorporates interdependent relationships between elements. Results illustrated that extensive technologies, constructed wetlands and pond systems are the most preferred alternatives by WWT experts. The sensitivity analysis performed verified that the ranking of WWT alternatives…

Pollutant removal efficiency of a pilot-scale Horizontal Subsurface Flow in Sicily (Italy) planted with Cyperus alternifolius L. and Typha latifolia …

2016

Constructed wetland systems (CWs) are developed biological technologies for the treatment and reuse of wastewater. The aims of this study were to evaluate the treatment performance of a pilot Horizontal Subsurface Flow system (HSSFs), to evaluate the reuse of treated wastewater (TWW) from CWs for the irrigation of giant reed (Arundo donax (L.)) and to assess the effects of TWW on the biomass yield of giant reed grown for pellet production. The research was carried out in Sicily (Italy) in a pilot-scale HSSFs which was fed with urban-treated urban wastewater following secondary treatment from an activated sludge wastewater treatment plant. Giant reed plots were irrigated with TWW from plante…
