1. Active prothoracic glands and oenocytes of last larval stage are both characteristized by well-developed smooth and rough endoplasmic reticulum (ER). Prothoracic glands also show plasma membrane infoldings, but not oenocytes which contain a large number of pleomorphic vesicles. 2. The fine structure of embryonic oenocytes corresponds after blastokinesis with that of active larval and adult cells. Thus, an activity in the late embryo can be assumed. Embryonic prothoracic glands reveal no signs of activity: smooth and rough ER are absent. The subcellular structure resembles that of organ anlagen, i.e. not yet fully differentiated tissue. The mode of growth of several tissues in Gryllus bimaculatus was investigated during postembryonic development by cytophotometric methods. In contrast to the situation in holo- metabolous insects, the tissues growing by endomitosis reach only moderate levels of polyploidy. In this case the growth of tissues is achieved by mitotic divisions of small cells with subsequent polyploidization. The time courses of DNA synthesis were measured within the 3rd and, for comparison, the 8th larval instar by incorporation of labelled thymidine followed by autoradiography. Using the Drosophila EcR-B1 cDNA as a probe, we have cloned the putative ecdysteroid receptor from the mealworm Tenebrio molitor. We have isolated two cDNAs with different 5' termini that contain a complete open reading frame. These two cDNAs encode two proteins with distinct N-terminal regions corresponding to two isoforms. The coleopteran receptor is obviously related to the ecdysteroid receptor of other insects, but shares only 89% and 61% amino acid identities with the DNA-binding and ligand-binding domains of the Drosophila receptor, respectively. 0962-1075 (Print) Journal Article Research Support, Non-U.S. Gov't; The accumulation of transcripts from two adult-specific cuticular genes (ACP-20 and ACP-22) is shown to be modified after addition of exogenous 20-hydroxyecdysone. In the continuous presence of high levels of the hormone, the expression of ACP-20 gene is significantly weaker than that of untreated controls, while ACP-22 expression is 2.5-fold increased. During active synthesis of the ACP messages, a 0.5 microg 20-hydroxyecdysone injection causes a rapid 2-fold increase in ACP-22 mRNA and is not able to repress ACP-20 mRNA accumulation. Individual measurements of ovarian cAMP concentrations and of ovarian biosynthesis of ecdysteroids, stimulated after a protein meal, demonstrated that steroidogenesis is preceded by a peak of cAMP in the ovaries. No genes have been identified that regulate growth by contributing to this key developmental decision to terminate feeding. The brain of Tenebrio molitor exhibited marked fluctuations in acetylcholinesterase (AChE) activity throughout metamorphosis. This was true AChE activity, since it was inhibited by high substrate concentrations and by 10 μM of the specific AChE inhibitor BW284C51 [(1,5-bis'4-allyldimethylammoniumphenyl)-pentan-3-one dibromide] but not by iso-OMPA (tetraisopropylpyrophosphoramide), a cholinesterase (but not AChE) inhibitor. Exogenous ecdysteroids (ecdysone, 20-hydroxyecdysone and makisterone A) administered to reproducing females of Oncopelus fasciatus (Dallas) reduced egg hatch drastically. The ovicidal activity apparently stemmed from ecdysteroids—unchanged hormone or metabolites—transferred from mother into oocyte. Different stages of embryogenesis were affected. Makisterone A, the predominant ecdysteroid in normal embryos, is by far the most effective ecdysteroid.

1. 1. Female-specific peptides can be detected by SDS-PAGE in the haemolymph when the female insects are 48 hr old. 2. 2. The age of the male insect at the moment of treatment with juvenile hormone I (JH I), JH III or ecdysone has no influence on the induction of the female-specific bands in male S. pandurus. 3. 3. Male insects exposed to the hormones for periods of time longer than 24 hr were also unable to show female-specific bands in their haemolymph. Consecutive treatments with JH were unable to induce these peptides in the males.
