Generation of oxysterols formed by free radicals and enzymes by electrochemical oxidation

International audience; It is commonly accepted that cholesterol oxide derivatives, also named oxysterols, are 27 carbon-atom molecules deriving either from enzymatic and non-enzymatic oxidation of cholesterol. Most of these compounds can be synthesized by more or less difficult and time consuming chemical reactions, and some of them have been discovered before the identification of the enzymes [mainly cytochrome P450 enzymes (CYP enzymes)] involved in their biosynthesis. A wide range of biological activities depends on oxysterols. Some oxysterols are also involved in the synthesis of cholesterol metabolites which have various properties. Antiinflammatory Therapy with Canakinumab for Atherosclerotic Disease

Background: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. Methods: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. AbstractGenetic and genomic architectures of traits under selection are key factors influencing evolutionary responses. Yet, knowledge of their impacts has been limited by a widespread assumption that most traits are controlled by unlinked polygenic architectures. Recent advances in genome sequencing and eco-evolutionary modelling are unlocking the potential for integrating genomic information into predictions of population responses to environmental change. Composition of the gut microbiota changes during ageing, but questions remain about whether age is also associated with deficits in microbiome function and whether these changes occur sharply or progressively.
