The Sea Urchin sns5 Insulator Protects Retroviral Vectors From Chromosomal Position Effects by Maintaining Active Chromatin Structure

Silencing and position-effect (PE) variegation (PEV), which is due to integration of viral vectors in heterochromatin regions, are considered significant obstacles to obtaining a consistent level of transgene expression in gene therapy. The inclusion of chromatin insulators into vectors has been proposed to counteract this position-dependent variegation of transgene expression. Here, we show that the sea urchin chromatin insulator, sns5, protects a recombinant gamma-retroviral vector from the negative influence of chromatin in erythroid milieu. Abstract The response to hyperosmotic stress is mediated by the HOG pathway. The MAP kinase Hog1 activates several transcription factors, regulates chromatin-modifying enzymes and, through its interaction with RNA polymerase II, it directs this enzyme to osmotic stress-controlled genes. For such targeting, this kinase requires the interaction with transcription factors Hot1 and Sko1. However, phosphorylation of these proteins by Hog1 is not required for their functionality. The DrosDel Deletion Collection: A Drosophila Genomewide Chromosomal Deficiency Resource

AbstractWe describe a second-generation deficiency kit for Drosophila melanogaster composed of molecularly mapped deletions on an isogenic background, covering ∼77% of the Release 5.1 genome. Using a previously reported collection of FRT-bearing P-element insertions, we have generated 655 new deletions and verified a set of 209 deletion-bearing fly stocks. In addition to deletions, we demonstrate how the P elements may also be used to generate a set of custom inversions and duplications, particularly useful for balancing difficult regions of the genome carrying haplo-insufficient loci. Contact-induced phenomena in the Alps

The main question underlying this chapter is to what extent language contact can affect syntactic structure. To tackle this issue we examine two relevant phenomena found in two minority languages spoken in the region Trentino-Alto Adige/South Tyrol: clitic climbing in Dolomitic Ladin and the use of the Romance complementizer ke in Cimbrian. Both phenomena are usually considered as the result of a contact-induced change influenced by the neighbouring Italo-Romance varieties. However, it is shown that the rising of clitic climbing is a language-internal process which is only accelerated by the contact with Italian. Virulence-Associated Mobile Elements in Bacilli and Clostridia

This chapter focuses on (i) species that induce human diseases, (ii) species that are able to produce toxins, and (iii) the association of appropriate virulence factors with possible mobile elements. With reference to bacilli, the chapter discusses mainly Bacillus anthracis and B. cereus. A section on clostridia focuses on Clostridium perfringens, neurotoxin-producing clostridia, and species capable of producing large clostridial cytotoxins (LCTs). The impact of the Little ice age on coccolithophores in the central Mediterranea Sea

The Little Ice Age (LIA) is the last episode of a series of Holocene climatic anomalies. There is still little knowledge on the response of the marine environment to the pronounced cooling of the LIA and to the transition towards the 20th century global warming. Here we present decadal-scale coccolithophore data from four short cores recovered from the central Mediterranean Sea (northern Sicily Channel and Tyrrhenian Sea), which on the basis of <sup>210</sup>Pb activity span the last 200–350 years. Goalkeeper: A Zero-Sum Exergame for Motivating Physical Activity

Incentives and peer competition have so far been employed independently for increasing physical activity. In this paper, we introduce Goalkeeper, a mobile application that utilizes deposit contracts for motivating physical activity in group settings. Goalkeeper enables one to set up a physical exercise challenge with a group of peers that deposit a fixed amount of money for participating. If a peer fails to complete the challenge, Goalkeeper redistributes their deposit to those who managed to complete it (i.e., zero-sum game).
