Reproducibility of Skin Temperature Response after Cold Stress Test Using the Game Ready System: Preliminary Study
2021
The objective of this preliminary study was to determine the reproducibility of lower limbs skin temperature after cold stress test using the Game Ready system. Skin temperature of fourteen participants was measured before and after cold stress test using the Game Ready system and it was repeated the protocol in four times: at 9:00, at 11:00, at 19:00, and at 9:00 h of the posterior day. To assess skin temperature recovery after cold stress test, a logarithmic equation for each region was calculated, and constant (β0) and slope (β1) coefficients were obtained. Intraclass correlation coefficient (ICC), standard error (SE), and within-subject coefficient of variation (CV) were determined. No …
No influence of magnetic fields on cell cycle progression using conditions relevant for patients during MRI.
2003
The purpose of this study was to examine whether exposure to magnetic fields (MFs) relevant for magnetic resonance imaging (MRI) in clinical routine influences cell cycle progression in two tumor cell lines in vitro. HL60 and EA2 cells were exposed to four types of MFs: (i) static MF of 1.5 and 7.05 T, (ii) extremely low frequency magnetic gradient fields (ELFMGFs) with ± 10 mT/m and 100 Hz, as well as ± 100 mT/m and 100 Hz, (iii) pulsed high frequency MF in the radiofrequency (RF) range (63.6 MHz, 5.8 μT), and (iv) a combination of (i–iii). Exposure periods ranged from 1 to 24 h. Cell cycle distribution (G0/G1, S, and G2/M phases) was analyzed by flow cytometry. Cell cycle analysis did not…
Hapten Design and Antibody Generation for Immunoanalysis of Spirotetramat and Spirotetramat-enol
2018
Spirotetramat - a tetramic acid insecticide - is rapidly metabolized or degraded to give spirotetramat-enol; so, common residue definitions include the sum of both compounds. In the present study, two spirotetramat-functionalized derivatives (haptens) have been designed to generate immunoreagents to these molecules for rapid immunochemical analysis. Haptens have been synthesized with alternative linker tethering sites and, for the first time, high-affinity antibodies have been generated with different specificities to these active principles. Two sensitive assays have been developed using the same antibody in different formats, and by using linker-site heterologous haptens, the selectivity …
Assessment of oral hygiene habits, oral hygiene practices and tooth wear among fertilizer factory workers of Northern India: A Cross sectional study
2015
Background: The association between oral hygiene habits & practices and severity of tooth wear lesion varies from community to community and also from occupation to occupation. The present study was conducted with to assess oral hygiene habits & practices and tooth wear among fertilizer factory workers of Punjab, India. Material and Methods: A descriptive cross sectional survey was conducted among 965 male workers who were aged between 19–58 years, who were the workers of fertilizers factory of Bathinda, India. An interview on the demographic profile, oral hygiene practices, and adverse habits followed a clinical examination for recording the Tooth Wear (Smith and Knight Index 1984) using T…
Reliability of a decision-tree model in predicting occupational lead poisoning in a group of highly exposed workers
2016
Objective This study aimed to provide the toxicological profile of some lead-exposed workers and obtain a predictive model for lead poisoning. Methods Data regarding external and absorbed exposure were collected from 585 subjects employed in ten metallurgical production departments. Airborne lead concentration, blood lead level (BLL), cumulative blood lead index (CBLI), urine delta-aminolevulinic acid (DALA), age, workplace/section, exposure period, and whether reported lead poisoning as occupational disease were examined using ANOVA, and, post-ANOVA, Pearson correlation matrix, PCA (principal component analysis), decision-tree modeling, and logistic modeling. Results BLL was less sensitive…
Rationally designed haptens for highly sensitive monoclonal antibody-based immunoanalysis of fenhexamid.
2018
Immunochemical methods have been consolidated during the last few years as complementary analytical strategies for chemical contaminant and residue determination. However, generation of suitable immunoreagents for small organic molecules demands adequate hapten design. In this study, fenhexamid was considered as a model compound and novel haptens were designed and synthesized in order to evaluate the influence of the linker tethering site on antibody binding properties and immunoassay parameters. Haptens were conceived with the spacer arm at different positions, while the more antigenic aromatic moiety was kept free. The synthesis of these functionalized compounds was accomplished by total …
Generation of anti-azoxystrobin monoclonal antibodies from regioisomeric haptens functionalized at selected sites and development of indirect competi…
2012
Azoxystrobin is a modern strobilurin fungicide used around the world to combat prime diseases affecting highly valuable crops. Accordingly, residues of this chemical are frequently found in food, even though mostly under maximum tolerated levels. We herein describe the development of an indirect competitive immunoassay for the determination of azoxystrobin residues. A panel of monoclonal antibodies displaying subnanomolar affinity to azoxystrobin was generated using, as immunizing haptens in mice, four functionalized derivatives carrying the same spacer arm located at different rationally chosen positions. This collection of antibodies was thoroughly characterized with homologous and hetero…
The role of the Thomsen-Friedenreich antigen as a tumor-associated molecule.
1990
The Thomsen-Friedenreich antigen (Gal-GalNAc) represents a tumor-associated molecule, which is assumed to be one of the few chemically well-defined antigens with a proven association with malignancy. In order to analyze the role of the carbohydrate structure Gal-GalNAc for gastrointestinal tumors, we immunized Balb/c mice with MCF-7 breast tumor cells together with synthetic Gal-GalNAc linked to a BSA carrier. One monoclonal antibody (82-A6) was established which recognizes the Thomsen-Friedenreich antigen according to the biochemical and serological analysis presented here. In contrast to the studies performed in the past, immunohistochemical results using this antibody 82-A6 did not exhib…
The Efflux Pump MexXY/OprM Contributes to the Tolerance and Acquired Resistance of Pseudomonas aeruginosa to Colistin
2020
The intrinsic resistance of Pseudomonas aeruginosa to polymyxins in part relies on the addition of 4-amino-4-deoxy-l-arabinose (Ara4N) molecules to the lipid A of lipopolysaccharide (LPS), through induction of operon arnBCADTEF-ugd (arn) expression. As demonstrated previously, at least three two-component regulatory systems (PmrAB, ParRS, and CprRS) are able to upregulate this operon when bacteria are exposed to colistin. In the present study, gene deletion experiments with the bioluminescent strain PAO1::lux showed that ParRS is a key element in the tolerance of P. aeruginosa to this last-resort antibiotic (i.e., resistance to early drug killing). Other loci of the ParR regulon, such as th…
In utero exposure to mixtures of xenoestrogens and child neuropsychological development.
2014
BACKGROUND: To date, no epidemiological studies have explored the impact and persistence of in utero exposure to mixtures of xenoestrogens on the developing brain. We aimed to assess whether the cumulative effect of xenoestrogens in the placenta is associated with altered infant neuropsychological functioning at two and at four years of age, and if associations differ among boys and girls. METHODS: Cumulative prenatal exposure to xenoestrogens was quantified in the placenta using the biomarker Total Effective Xenoestrogen Burden (TEXB-alpha) in 489 participants from the INMA (Childhood and the Environment) Project. TEXB-alpha was split in tertiles to test its association with the mental and…
