Steroids are used as faecal markers in environmental and in archaeological studies, because they provide insights into ancient agricultural practices and the former presence of animals. Up to now, steroid analyses could only identify and distinguish between herbivore, pig, and human faecal matter and their residues in soils and sediments. We hypothesized that a finer differentiation between faeces of different livestock animals could be achieved when the analyses of several steroids is combined (Δ5-sterols, 5α-stanols, 5β-stanols, epi-5β-stanols, stanones, and bile acids). There is a lack of biomarkers or indices that can be used to predict the quality of fish semen samples following the freezing and thawing cycle. In the present study, a series of semen indices were tested to assess if they could accurately forecast the cryopreservation potential of Atlantic cod (Gadus morhua) semen. Fresh and frozen-thawed sperm activity variables were compared, and relationships between frozen-thawed sperm activity and fertilization success were examined. In comparison with fresh sperm, activity variables of frozen-thawed spermatozoa were reduced. A specific pressure-assisted CE-MS method is described for the analysis of five quinolone residues. MS using a single quadrupole is compared with multiple-stage MS using a quadrupole IT (QIT-MS(n)). The procedure involves a common sample preparation by SPE on disposable cartridges. The most suitable electrolyte is 60 mM (NH(4))(2)CO(3) at pH 9.2. Single quadrupole does not provide enough fragmentation to confirm identities according to the current legislation. However, QIT-MS(n) achieves selective fragmentation.
