An efficient Escherichia coli expression system for the production of a functional N-terminal domain of the T1R3 taste receptor.

2012

http://www.landesbioscience.com/; International audience; Sweet taste is mediated by a dimeric receptor composed of two distinct subunits, T1R2 and T1R3, whereas the T1R1/T1R3 receptor is involved in umami taste perception. The T1R1, T1R2, and T1R3 subunits are members of the small family of class C G protein-coupled receptors (GPCRs). The members of this family are characterized by a large N-terminal domain (NTD), which is structurally similar to bacterial periplasmic-binding proteins and contains the primary ligand-binding site. In a recent study, we described a strategy to produce a functional dimeric human T1R3-NTD. Although the protein was expressed as inclusion bodies (IBs) using the …

Bleomycin, a selective inhibitor of DNA-dependent DNA polymerase from oncogenic RNA viruses.

1972

Abstract Bleomycin, an antibiotic, inhibits the DNA-dependent DNA polymerase from Rauscher murine leukemia virus. Higher concentrations of BLM ∗ are required to inhibit it's RNA-dependent DNA polymerase. These inhibition effects of the non-competitive type are not altered by preincubation of the DNA with BLM. Under comparable conditions neither the DNA-dependent DNA polymerase activity from E. coli and mouse liver nor the DNA-dependent RNA polymerase activity from mouse lymphoma cells are affected by BLM.

Esimerkiksi The Walking Dead -sarjan ensimmäinen jakso näytettiin samanaikaisesti 120 maassa, ja sarjaa on sittemmin katsonut tuotantokaudesta riippuen 5–15 miljoonaa katsojaa. Post-apokalyptinen maailma kiehtoo suurta yleisöä, koska dystooppiset tarinat peilaavat erilaisia käsityksiä ihmisten perusolemuksesta ja yhteiskunnan perustasta; sarjaa voi pitää filosofisena ajatuskokeena, jolla testataan, miten erilaiset näkemykset toimisivat luonnontilaan paluun jälkeen. Artikkelimme lähtökohta on, että zombikuvasto tarjoaa hedelmällisen tilaisuuden filosofiselle analyysille. Tarkastelemme yhteisku…

Regularization Preserving Localization of Close Edges

2007

International audience; In this letter, we address the problem of the influence of neighbor edges and their effect on the edge delocalization while extracting a neighbor contour by a derivative approach. The properties to be fulfilled by the regularization operators to minimize or suppress this side effect are deduced, and the best detectors are pointed out. The study is carried out in 1-D for discrete signal. We show that among the derivative filters, one of them can correctly detect our model edges without being influenced by a neighboring transition, whatever their separation distance is and their respective amplitude is. A model of contour and close transitions is presented and used through…

Functional interactions between members of the REPAT family of insect pathogen-induced proteins

2012

Studies on the transcriptional response to pathogens in the insect larval gut have shown the regulation of several genes after the infection. Repat (REsponse to PAThogens) genes were first identified in Spodoptera exigua midgut as being up-regulated in response to the exposure to Bacillus thuringiensis toxins and baculovirus. Recently, new members of the REPAT family showed a constitutive up-regulation in a B. thuringiensis-resistant population. Based on a yeast two-hybrid screening, we have detected the interaction of REPAT1 with other members of the REPAT family, leading to the discovery of a new member: REPAT8. The functional role of this interaction was shown by following the changes of…

L‐Aspartate as a high‐quality nitrogen source in Escherichia coli : Regulation of L‐aspartase by the nitrogen regulatory system and interaction of L‐…

2020

Escherichia coli uses the C4-dicarboxylate transporter DcuA for L-aspartate/fumarate antiport, which results in the exploitation of L-aspartate for fumarate respiration under anaerobic conditions and for nitrogen assimilation under aerobic and anaerobic conditions. L-Aspartate represents a high-quality nitrogen source for assimilation. Nitrogen assimilation from L-aspartate required DcuA, and aspartase AspA to release ammonia. Ammonia is able to provide by established pathways the complete set of intracellular precursors (ammonia, L-aspartate, L-glutamate, and L-glutamine) for synthesizing amino acids, nucleotides, and amino sugars. AspA was regulated by a central regulator of nitrogen meta…

Expression, purification, crystallization and preliminary X-ray analysis of perakine reductase, a new member of the aldo-keto reductase enzyme superf…

2006

Perakine reductase (PR) is a novel member of the aldo-keto reductase enzyme superfamily from higher plants. PR from the plant Rauvolfia serpentina is involved in the biosynthesis of monoterpenoid indole alkaloids by performing NADPH-dependent reduction of perakine, yielding raucaffrinoline. However, PR can also reduce cinnamic aldehyde and some of its derivatives. After heterologous expression of a triple mutant of PR in Escherichia coli, crystals of the purified and methylated enzyme were obtained by the hanging-drop vapour-diffusion technique at 293 K with 100 mM sodium citrate pH 5.6 and 27% PEG 4000 as precipitant. Crystals belong to space group C222(1) and diffract to 2.0 A, with unit-…

Control of Growth and Persistence of Listeria monocytogenes and β-Lactam-Resistant Escherichia coli by Thymol in Food Processing Settings

2020

The main objective of this study was to evaluate the efficacy of thymol in controlling environmental contamination in food processing facilities. The effect of thymol was tested as an agent to prevent planktonic and bacterial biofilm growth of twenty-five Listeria monocytogenes isolates from a variety of foods and five Escherichia coli isolates from a farm. The E. coli isolates were positive for extended spectrum &beta
