Protein sorting in Plasmodium falciparum-infected red blood cells permeabilized with the pore-forming protein streptolysin O

1996

Plasmodium falciparum is an intracellular parasite of human red blood cells (RBCs). Like many other intracellular parasites, P. falciparum resides and develops within a parasitophorous vacuole which is bound by a membrane that separates the host cell cytoplasm from the parasite surface. Some parasite proteins are secreted into the vacuolar space and others are secreted, by an as yet poorly defined pathway, into the RBC cytosol. The transport of proteins from the parasite has been followed mainly using morphological methods. In search of an experimental system that would allow (i) dissection of the individual steps involved in transport from the parasite surface into the RBC cytosol, and (ii…

Organochlorine Pesticides and Polychlorinated Biphenyl Congeners in LannerFalco biarmicus feldeggiiSchlegel Chicks and Lanner Prey in Sicily, Italy

2008

This paper reports on research conducted to elucidate the risk posed to the Sicilian population of the endangered lanner falcon Falco biarmicus feldeggii Schlegel by organochlorine (OC) pesticides and polychlorinated biphenyls (PCBs), as part of a wider study on contaminant risk to the lanner. Seventeen lanner nest sites were studied in northern and central Sicily. Sampling (in 2005) and analysis were carried out for selected OC pesticides and PCB congeners in lanner chick blood (15 chicks from 6 nest sites) and in two of the main lanner prey species, magpie Pica pica (36 individuals from 6 lanner nest sites) and rock dove Columba livia (10 individuals from 2 lanner nest sites). No OC and P…

Landscape-scale Spatial Distribution of the Lanner Falcon (Falco biarmicus feldeggii) Breeding Population in Italy

2008

Falco biarmicus feldeggii is one of the most threatened taxa in Europe. Its global population is estimated at a few hundred pairs unequally scattered in a vast and fragmented area stretching from Sicily to the Caspian Sea. Most recent counts showed that Italy hosts a large part (>25%) of the whole population. Consequently, Italian authorities promoted a national action plan. In this framework, we carried out the first national survey for the Lanner Falcon in Italy (2003-2004). Our study area covered the whole breeding range, i.e., Sicily and the Italian peninsula (n = 2909 cells 10 × 10 km). When possible, we considered also additional information from previous regional investigations (1993…

Long-lived contrails and convective cirrus above the tropical tropopause

2017

Abstract. This study has two objectives: (1) it characterizes contrails at very low temperatures and (2) it discusses convective cirrus in which the contrails occurred. (1) Long-lived contrails and cirrus from overshooting convection are investigated above the tropical tropopause at low temperatures down to −88 °C from measurements with the Russian high-altitude research aircraft M-55 Geophysica, as well as related observations during the SCOUT-O3 field experiment near Darwin, Australia, in 2005. A contrail was observed to persist below ice saturation at low temperatures and low turbulence in the stratosphere for nearly 1 h. The contrail occurred downwind of the decaying convective system H…

Python valodas REST API ietvaru salīdzinājums

2016

Polyoxygenated Cyclohexenes and Other Constituents of Cleistochlamys kirkii Leaves.

2016

Thirteen new metabolites, including the polyoxygenated cyclohexene derivatives cleistodiendiol (1), cleistodienol B (3), cleistenechlorohydrins A (4) and B (5), cleistenediols A-F (6-11), cleistenonal (12), and the butenolide cleistanolate (13), 2,5-dihydroxybenzyl benzoate (cleistophenolide, 14), and eight known compounds (2, 15-21) were isolated from a MeOH extract of the leaves of Cleistochlamys kirkii. The purified metabolites were identified by NMR spectroscopic and mass spectrometric analyses, whereas the absolute configurations of compounds 1, 17, and 19 were established by single-crystal X-ray diffraction. The configuration of the exocyclic double bond of compound 2 was revised base…

Use of poly(amidoamine) drug conjugates for the delivery of antimalarials to Plasmodium

2013

Current malaria therapeutics demands strategies able to selectively deliver drugs to Plasmodium-infected red blood cells (pRBCs) in order to limit the appearance of parasite resistance. Here, the poly(amidoamines) AGMA1 and ISA23 have been explored for the delivery of antimalarial drugs to pRBCs. AGMA1 has antimalarial activity per se as shown by its inhibition of the in vitro growth of Plasmodium falciparum, with an IC50 of 13.7 μM. Fluorescence-assisted cell sorting data and confocal fluorescence microscopy and transmission electron microscopy images indicate that both polymers exhibit preferential binding to and internalization into pRBCs versus RBCs, and subcellular targeting to the par…

Molecular interaction of artemisinin with translationally controlled tumor protein (TCTP) of Plasmodium falciparum

2012

Malaria causes millions of death cases per year. Since Plasmodium falciparum rapidly develops drug resistance, it is of high importance to investigate potential drug targets which may lead to novel rational therapy approaches. Here we report on the interaction of translationally controlled tumor protein of P. falciparum (PfTCTP) with the anti-malarial drug artemisinin. Furthermore, we investigated the crystal structure of PfTCTP. Using mass spectrometry, bioinformatic approaches and surface plasmon resonance spectroscopy, we identified novel binding sites of artemisinin which are in direct neighborhood to amino acids 19-46, 108-134 and 140-163. The regions covered by these residues are know…
