ALICE forward rapidity upgrades

Two forward rapidity upgrades of the ALICE apparatus at the LHC are presented: the Muon Forward Tracker (MFT) and the Fast Interaction Trigger (FIT). Their designs are driven by physics performance studies, which have been done using MC simulations. Several prototypes have been tested. Currently the detector design phase has ended. The final layouts, geometries and expected performance figures are presented. The latest developments from the test of the prototypes are discussed. The MFT will improve the performance of the ALICE Muon Spectrometer by adding vertexing capabilities to the system. Unusual internal structure of cm-sized coldwater calcite: Weichselian spars in former pools of the Zinnbergschacht Cave (Franconian Alb/SE Germany)

The investigation of the internal structure of calcite crystals is a new focus in speleothem science, especially in the range of crystallization temperatures close to 0°C. Recently found calcite spars from Zinnbergschacht Cave of the Franconian Alb (SE Germany) are ideal for multi-method investigation. The elongated calcites (up to 6 cm in length) with three to six lateral faces and basal triangular faces at the ends are observed in collapse-zones in the cave. 230Th/U-ages of 38.9 ka suggest formation during the periglacial Weichselian, between the Scandinavian and Alpine Glaciations. The novel NF-κB inhibitor DHMEQ synergizes with celecoxib to exert antitumor effects on human liver cancer cells by a ROS-dependent mechanism

In a previous work of ours dehydroxymethyl-epoxyquinomicin (DHMEQ), an inhibitor of NF-κB, was shown to induce apoptosis through Reactive Oxygen Species (ROS) production in hepatoma cells. The present study demonstrated that DHMEQ cooperates with Celecoxib (CLX) to decrease NF-κB DNA binding and to inhibit cell growth and proliferation more effectively than treatment with these single agents alone in the hepatoma cell lines HA22T/VGH and Huh-6. ROS production induced by the DHMEQ-CLX combination in turn generated the expression of genes involved in endoplasmic reticulum (ER) stress and silencing TRB3 mRNA significantly decreased DHMEQ-CLX-induced cell growth inhibition. SAHA induces apoptosis in hepatoma cells and synergistically interacts with the proteasome inhibitor Bortezomib.

Histone deacetylase (HDAC) inhibitors represent a promising group of anticancer agents. This paper shows that the HDAC inhibitor suberoylanilide hydroxamic acid (SAHA) stimulated at 5-10 microM apoptosis in human hepatoma HepG2 and Huh6 cells, but was ineffective in primary human hepatocytes (PHH). In HepG2 cells SAHA induced the extrinsic apoptotic pathway, increasing the expression of both FasL and FasL receptor and causing the activation of caspase-8. Membrane vesicles shed by oligodendroglioma cells induce neuronal apoptosis.

In order to investigate the mechanism by which oligodendrogliomas cause neuronal damage, media conditioned by G26/24 oligodendroglioma cells, were fractionated into shed vesicles and vesicle-free supernatants, and added to primary cultures of rat fetal cortical neurons. After one night treatment with vesicles, a reproducible, dose-dependent, inhibitory effect on neurite outgrowth was already induced and, after 48-72 h of incubation, neuronal apoptosis was evident. Vesicle-free supernatants and vesicles shed by NIH-3T3 cells had no inhibitory effects on neurons. Ultraviolet light-induced DNA damage triggers apoptosis in nucleotide excision repair-deficient cells via Bcl-2 decline and caspase-3/-8 activation.

Ultraviolet (UV) light is a potent mutagenic and genotoxic agent. Whereas DNA damage induced by UV light is known to be responsible for UV-induced genotoxicity, its role in triggering apoptosis is still unclear. We addressed this issue by comparing nucleotide excision repair (NER) deficient 27-1 and 43-3B Chinese hamster (CHO) cells with the corresponding wild-type and ERCC-1 complemented cells. It is shown that NER deficient cells are dramatically hypersensitive to UV-C induced apoptosis, indicating that DNA damage is the major stimulus for the apoptotic response.
