Muscarinic M2 receptors in acetylcholine-isoproterenol functional antagonism in human isolated bronchus

The muscarinic functional antagonism of isoproterenol relaxation and the contribution of muscarinic M2 receptors were examined in human isolated bronchus. In intact tissues, acetylcholine (ACh) precontraction decreased isoproterenol potency and maximal relaxation (−log EC50 shift = −1.49 ± 0.16 and Emax inhibition for 100 μM ACh = 30%) more than the same levels of histamine contraction. The M2receptor-selective antagonist methoctramine (1 μM) reduced this antagonism in ACh- but not histamine-contracted tissues. Similar results were obtained for forskolin-induced relaxation. Baltais fosfors ir molekulārs kristāls, kas sastāv no tetraedra formas P4 molekulām, kuru savstarpējais izkārtojums kristāliskajā režģī mainās, pārejot starp trim pazīstamajām fāzēm: α; β; γ. Materiāls ir pazīstams jau sen, taču kā molekulārs kristāls tas ir pētīts salīdzinoši maz, it īpaši ar svārstību spektroskopijas metodēm, kas, piemēram, spētu apstiprināt vai noliegt no rentgenstaru difrakcijas iegūto kristālisko struktūru pareizību. Šajā darbā eksperimentāli iegūti baltā fosfora α un β fāzu Ramana izkliedes spektri pie dažādām temperatūrām apgabalā no -125°C līdz -20°C. Raman spectroscopic data were obtained for (Mg,Fe)2SiO4 samples during compression to 57 GPa. Single crystals of San Carlos olivine compressed hydrostatically above 41 GPa showed appearance of a new “defect” peak in the 820–840 cm−1 region associated with SiOSi linkages appearing between adjacent Embedded Image tetrahedra to result in five- or sixfold-coordinated silicate species. Appearance of this feature is accompanied by a broad amorphous background. Acetylcholine (ACh) acts as a local cellular signaling molecule and is widely expressed in nature, including mammalian cells and embryonic stem cells. The murine embryonic stem cell line CGR8 synthesizes and releases substantial amounts of ACh. Particularly during early differentiation - a period associated with multiple alterations in geno-/phenotype functions - synthesis and release of ACh are increased by 10-fold. In murine stem cells second messengers of the STAT-3, PI3K and cAMP/PKA pathways are involved in maintaining self-renewal and pluripotency.
