Xrcc2 deficiency sensitizes cells to apoptosis by MNNG and the alkylating anticancer drugs temozolomide, fotemustine and mafosfamide

DNA double-strand breaks (DSBs) are potent killing lesions, and inefficient repair of DSBs does not only lead to cell death but also to genomic instability and tumorigenesis. DSBs are repaired by non-homologous end-joining and homologous recombination (HR). A key player in HR is Xrcc2, a Rad51-like protein. Cells deficient in Xrcc2 are hypersensitive to X-rays and mitomycin C and display increased chromosomal aberration frequencies. In order to elucidate the role of Xrcc2 in resistance to anticancer drugs, we compared Xrcc2 knockout (Xrcc2-/-) mouse embryonic fibroblasts with the corresponding isogenic wild-type and Xrcc2 complemented knockout cells. The DNA repair protein O(6)-methylguanine-DNA methyltransferase (MGMT) is an important suicide enzyme involved in the defense against O(6)-alkylating mutagens. It also plays a role in the resistance of tumors to anticancer drugs targeting the O(6)-position of guanine, such as temozolomide and fotemustine. Several potent MGMT inhibitors have been developed sensitizing cells to O(6)-alkylating agents. Aimed at targeting MGMT inhibitors to tumor cells, we synthesized MGMT inhibitory compounds conjugated with glucose to improve uptake in tumor cells. Sono, in particolare, evidenziati il metodo diretto e il metodo fotogrammetrico che, congiuntamente, permettono di ottimizzare la rappresentazione dell'architettura storica in vista delle elaborazioni grafiche del progetto. The poster concerns the metric survey of historic architecture for the restoration project. Are, in particular, highlighted the direct method and the photogrammetric method, which, together, allow you to optimize the representation of ancient architecture.

Multidrug and broad-spectrum cephalosporin resistance among Salmonella enterica serotype enteritidis clinical isolates in southern Italy.

ABSTRACT From 1992 to 1997, only six sporadic isolates of Salmonella enterica serotype Enteritidis from patients with cases of gastroenteritis in southern Italy exhibited resistance to broad-spectrum cephalosporins. Five isolates produced SHV-12, and one isolate encoded a class C β-lactamase. The bla SHV-12 gene was located in at least two different self-transferable plasmids, one of which also carried a novel class 1 integron.

Dissemination of CTX-M-Producing Escherichia coli in Freshwater Fishes From a French Watershed (Burgundy)

International audience; The burden of extended-spectrum beta-lactamases producing Escherichia coli (ESBL-Ec), has increased over several decades. Freshwater ecosystems are suspected to play an important ecological and evolutionary role in driving the dissemination of antimicrobial resistance. The aim of our study was to decipher the occurrence of ESBL-Ec in a small watershed (Ouche river, Burgundy, France), targeting environmental matrices and fishes. [EN] Metric and graphic documentation of archaeological cultural heritage requires effective solutions for archiving, visualization, analysis, management, and monitoring of different monuments and sites. This paper presents the geomatic workflow undertaken on a singular rock art site, Coves de la Saltadora. The cave is part of the Rock Art of the Mediterranean Basin on the Iberian Peninsula declared World Heritage Site by UNESCO in1998. The different steps are reviewed, starting from the data acquisition until the final delivery of part of the metric outputs such as 3D models, photorealistic models, orthophoto mosaics in two levels, general and detail.

Site-by-site tracking of signal transduction in an azidophenylalanine-labeled bacteriophytochrome with step-scan FTIR spectroscopy

Signal propagation in photosensory proteins is a complex and multidimensional event. Unraveling such mechanisms site-specifically in real time is an eligible but a challenging goal. Here, we elucidate the site-specific events in a red-light sensing phytochrome using the unnatural amino acid azidophenylalanine, vibrationally distinguishable from all other protein signals. In canonical phytochromes, signal transduction starts with isomerization of an excited bilin chromophore, initiating a multitude of processes in the photosensory unit of the protein, which eventually control the biochemical activity of the output domain, nanometers away from the chromophore.
