Recurrence of pleomorphic adenoma of the parotid gland-predictive value of cadherin-11 and fascin
2008
The predictive value of cadherin-11, tenascin, fascin, and mucin-1 as markers for the likelihood of recurrence in pleomorphic adenoma of the parotid gland was examined. In this retrospective study we analysed 20 tumours from16 patients by immunohistochemistry. Staining intensities were measured using a semiquantitative scoring approach; localisation (tumour centre vs border) as well as clinical data were analysed and correlated with follow-up. Cadherin-11 was increased in recurrent tumours. However, no changes of fascin, tenascin or mucin-1 were observed. Cadherin-11 and fascin were increased in primary tumours of patients with later recurrence, with fascin upregulation restricted to the tu…
Fascin upregulation in primary head and neck squamous cell carcinoma is associated with lymphatic metastasis
2014
Fascin is an actin-bundling protein that is associated with cellular motility and cancer-cell invasion. The present study aimed to examine the expression of fascin in head and neck squamous cell carcinoma (HNSCC) and its potential use as a biomarker. In a prospective study with a median follow-up time of 48.8 months, tumor tissues, adjacent healthy tissues and cervical lymph node metastases were collected from 25 patients and analyzed by immunohistochemistry. The specimens were scored according to the intensity of fascin staining and the percentage of tumor cells stained using a semi-quantitative scoring approach; the data were analyzed and correlated with clinical follow-up observations. A…
The human fascin gene promoter is highly active in mature dendritic cells due to a stage-specific enhancer.
2003
Abstract Dendritic cells (DC), regarded as the most efficient APCs of the immune system, are capable of activating naive T cells. Thus, DC are primary targets in immunotherapy. However, little is known about gene regulation in DC, and for efficient transcriptional targeting of human DC, a suitable promoter is still missing. Recently, we successfully used the promoter of the murine actin-bundling protein fascin to transcriptionally target DC by DNA vaccination in mice. In this study, we report on isolation of the human fascin promoter and characterization of its regulatory elements. The actively expressed gene was distinguished from a conserved inactive genomic locus and a continuous region …
Effects of small interfering RNAs targeting fascin on human esophageal squamous cell carcinoma cell lines
2010
Abstract Background Fascin induces membrane protrusions and cell motility. Fascin overexpression was associated with poor prognosis, and its downregulation reduces cell motility and invasiveness in esophageal squamous cell carcinoma (ESCC). Using a stable knockdown cell line, we revealed the effect of fascin on cell growth, cell adhesion and tumor formation. Methods We examined whether fascin is a potential target in ESCC using in vitro and in vivo studies utilizing a specific siRNA. We established a stable transfectant with downregulated fascin from KYSE170 cell line. Results The fascin downregulated cell lines showed a slower growth pattern by 40.3% (p In vivo, the tumor size was signific…
Biallelic mutations in neurofascin cause neurodevelopmental impairment and peripheral demyelination
2019
See Karakaya and Wirth (doi:10.1093/brain/awz273) for a scientific commentary on this article. Neurofascin (NFASC) isoforms are immunoglobulin cell adhesion molecules involved in node of Ranvier assembly. Efthymiou et al. identify biallelic NFASC variants in ten unrelated patients with a neurodevelopmental disorder characterized by variable degrees of central and peripheral involvement. Abnormal expression of Nfasc155 is accompanied by severe loss of myelinated fibres.
A Literary Fascination with China: Three Contemporary Novels by 'Foreign' Writers.
2019
The paper is about a contemporary tendency among writers from non-Chinese countries to write novels that present deep connections with China. The authors taken into consideration are from Australia: Nicholas Jose and Gail Jones, and from Italy: Isaia Iannaccone.
Roma pagana, Roma cristiana, Roma nostra : l'urbe nella cultura umanistica polacca
2014
Rome was a crucial point of reference for the culture which was developing in the Polish State. An important role played the fascination with antiąuity and the attachment to Christian values inseparably linked to Rome. One of the examples, among others, was the creation of legendary stories about the relations between the aristocratic families of Poland and Lithuania and the Roman families front the Empire times. The conviction of the Polish nations mission becomes a cultural and morał means of civilization achievements having Rome in its centre.
Organic waste materials for Bioengineering works
2016
Bioengineering uses plant and biodegradable materials of natural origin, stones, steel, additives and synthetic products in various combinations and as support for the growth of plants. The lack of available resources and progressive increasing of desertification in Sicily, led to the search for alternative materials. Objective of the work is testing organic waste materials for the realization of bioengineering works in the several areas of application: terrestrial, fluvial and coastline. To this aim, is proposed the use of innovative techniques that involve the construction of low-cost brushwood, environmentally friendly materials made: the pruning of vines and the oceanic Posidonia oceani…
Expression of the actin-bundling protein fascin in cultured human dendritic cells correlates with dendritic morphology and cell differentiation.
2000
Dendritic cells are key players of the immune system as they efficiently induce primary immune responses by activating naive T cells. We generated human dendritic cells from CD14+ blood precursors and investigated expression of the actin-bundling protein fascin during maturation by western blotting, immunofluorescence, and cytofluorometry. Cells obtained by culture of CD14+ blood precursors in the presence of granulocyte-macrophage colony-stimulating factor and interleukin-4, which were only weakly positive for the maturation marker CD83, expressed low amounts of fascin. Addition of a cytokine cocktail including tumor necrosis factor alpha, interleukin-1beta, interleukin-6, and prostaglandi…
