A method for the quantitative analysis of chromite ores by x-ray fluorescence spectrometry using beads is proposed. The work concerned the serious problems caused by the refractory nature of these materials which prevents the use of glass discs in x-ray fluorescence. An in-depth study was done to optimise the variables which influence the glass disc formation process. Sufficiently homogeneous glass discs were obtained under the following experimental conditions: lithium tetraborate as flux with moderate sample dilution (1:40), with the addition of one or two drops of LiBr solution(250 g l−1), at a temperature of 1200 °C for 30 min. We report the observation of the redox state of the blue copper protein azurin on the single-molecule level. The fluorescence of a small fluorophore attached to the protein is modulated by the change in absorption of the copper center via fluorescence resonance energy transfer (FRET). In our model system, the fluorescence label Cy5 was coupled to azurin from Pseudomonas aeruginosa via cysteine K27C. The Cy5 fluorescence was partially quenched by the absorption of the copper center of azurin in its oxidized state. In the reduced state, absorption is negligible, and thus no quenching occurs. Abstract Melittin, a C-terminal peptide, incorporated the fluorescent probe monodansylcadaverine (DNC) when catalysed by guinea-pig liver transglutaminase and Ca2+, as determined by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC). A 1:1 adduct DNC-melittin was identified in which a single glutamine residue out of two, i.e. Gln25, acts as acyl donor. Incubation of melittin with transglutaminase in the absence of DNC originated high molecular mass complexes indicative that the peptide lysine residue can act as an acyl acceptor. AbstractEleven trace elements (Ba, Cu, Fe, Mn, Ni, Pb, Rb, Se, Sr, V, Zn) were quantitatively determined in hen egg samples collected from different poultry housing types (large-scale poultry farms, organic farms and domestic farms) over the territory of Latvia. Samples were wet digested and quantitatively analysed by total reflection X-ray fluorescence spectrometry (TXRF). A semipurified C. rugosa lipase (LS) has been prepared from commercial lipase (LC) using an economical procedure. The presence of sugars and glycopeptides has been detected in LS and LC. Pure lipase only has covalently bonded sugars. The hydrolysis of olive oil catalyzed by LS and commercial lipase (LC) is sensitive to the presence of cations Na(I), Mg(II), Ca(II), and Ba(II) and to the nature of buffer. This work summarizes the characterization of flint artefacts from the Middle Palaeolithic site ‘Abrigo de la Quebrada’ (Chelva, Valencia, Spain) and flint geological samples collected in the Chelva area. A HPLC-fluorescence method for characterization of proteinaceous glues from binding media used in pictorial works of art prior to conservation or restoration treatment is proposed. Fluorescence derivatization of amino acids released by acid hydrolysis of standard proteins is studied. The derivatization reagent was o-phtalaldehyde with 2-mercaptoethanol as catalyst. Mobile phase was a programmed gradient among two eluents (water buffered at pH 5.8 wit 5% THF, and methanol) and is able to satisfactorily resolve the amino acid derivatives in 45min. Abstract The micromorphological variations of amphiphilic L -glutamate derivatives at air-substrate interfaces due to an adjacent mobile polymer matrix have been investigated via fluorescence microscopy.
