Cytometric analysis for drug-induced steatosis in HepG2 cells

Drugs are capable of inducing hepatic lipid accumulation. When fat accumulates, lipids are primarily stored as triglycerides which results in steatosis and provides substrates for lipid peroxidation. An in vitro multiparametric flow cytometry assay was performed in HepG2 cells by using fluorescent probes to analyze cell viability (propidium iodide, PI), lipid accumulation (BODIPY493/503), mitochondrial membrane potential (tetramethyl rhodamine methyl ester, TMRM) and reactive oxygen species generation (ROS) (2',7'-dihydrochlorofluorescein diacetate, DHCF-DA) as functional markers. Multi frequency phase fluorimetry (MFPF) for oxygen partial pressure measurement: ex vivo validation by polarographic clark-type electrode.

BACKGROUND: Measurement of partial pressure of oxygen (PO2) at high temporal resolution remains a technological challenge. This study introduces a novel PO2 sensing technology based on Multi-Frequency Phase Fluorimetry (MFPF). The aim was to validate MFPF against polarographic Clark-type electrode (CTE) PO2 measurements. METHODOLOGY/PRINCIPAL FINDINGS: MFPF technology was first investigated in N = 8 anaesthetised pigs at FIO2 of 0.21, 0.4, 0.6, 0.8 and 1.0. The title compounds both exhibit strong features of interaction of their two lowest excited states, but only one of them, the dimethylamino derivative, exhibits dual fluorescence. This is a direct indication that state interaction and dual fluorescence are not directly related.

Intramolecular charge transfer and dual fluorescence of 4-(dimethylamino)benzonitrile: ultrafast branching followed by a two-fold decay mechanism.

International audience; In this contribution we present new experimental and theoretical results for the intramolecular charge transfer (ICT) reaction underlying the dual fluorescence of 4-(dimethylamino) benzonitrile ( DMABN), which indicate that the fully twisted ICT (TICT) state is responsible for the time-resolved transient absorption spectrum while a distinct partially twisted ICT (pTICT) structure is suggested for the fluorescent ICT state.

Ground and excited state dipole moments of coumarin laser dyes: Investigation by electro-optical absorption and emission methods

Abstract Modified electro-optical absorption and emission methods were used to measure the dipole moments of four efficient coumarin laser dyes (CU1, CU4, CU120, CU334) in the equilibrated ground, excited Franck-Condon and equilibrated excited states. The measurements were performed in cyclohexane (CU1, CU120, CU334) and dioxane (all CUs) at room temperature. Our results show that the charge distribution in CU4 differs substantially from that in the other CUs. The ground electronic state of KCs studied by Fourier transform spectroscopy

We present here the first analysis of laser induced fluorescence (LIF) of the KCs molecule obtaining highly accurate data and perform a direct potential construction for the X (1)Sigma(+) ground state in a wide range of internuclear distances. KCs molecules were produced by heating a mixture of K and Cs metals in a heat pipe at a temperature of about 270 degrees C. KCs fluorescence was induced by different laser sources: the 454.5, 457.9, 465.8, and 472.7 nm lines of an Ar(+) laser, a dye laser with Rhodamine 6G dye (excitation at around 16 870 cm(-1)), and 850 and 980 nm diode lasers (11 500-11 900 and 10 200-10 450 cm(-1) tuning ranges, respectively). Micellization properties of cardanol as a renewable co-surfactant

With the aim to improve the features of surfactant solutions in terms of sustainability and renewability we propose the use of hydrogenated natural and sustainable plant-derived cardanol as an additive to com- mercial surfactants. In the present study we demonstrated that its addition, in amounts as high as 10%, to commercial surfactants of different charge does not significantly affect surfactant properties. Conversely, the presence of hydrogenated cardanol can strongly affect spectrophotometric determination of CMC if preferential interactions with the dyes used take place. ENERGY DISPERSIVE X-RAY DIFFRACTION IN CULTURAL HERITAGE SCIENCE: THE WINNING DUO OF STRUCTURAL AND ELEMENTAL ANALYSIS

The capabilities of Energy-Dispersive X-ray Diffraction (EDXD) in the field of Cultural Heritage are disccussed. EDXD diffractometer, equipped with a white source, is particularly suited for the structural and elemental analysis of items having different nature. Given its compact and versatile design, the instrument allows to collect data from samples without tearing down of any portion and with no specific preparation. In the wide energy range sampled during the measurements, both X-ray fluorescence and diffraction features appear. In all cases, data are acquired in air and in a non-destructive way, and their collection is fast.

Automated analysis of images for molecular quantification in immunohistochemistry.

The quantification of the expression of different molecules is a key question in both basic and applied sciences. While protein quantification through molecular techniques leads to the loss of spatial information and resolution, immunohistochemistry is usually associated with time-consuming image analysis and human bias. In addition, the scarce automatic software analysis is often proprietary and expensive and relies on a fixed threshold binarization.
