The Na+-K+-2Cl- cotransporter blocker furosemide inhibits gamma-aminobutyric acid (GABA)-gated chloride currents and reverses GABA-mediated inhibition of [35S]-t-butylbicyclophosphorothionate ([35S]TBPS) binding of the cerebellar alpha6 subunit-containing GABA(A) receptors much more potently than the cerebrocortical non-alpha6 subunit-containing receptors. A pilot study demonstrating how inhaled furosemide enhances the bronchodilator effect of salbutamol

Abstract To investigate whether inhaled furosemide can improve the bronchodilator effect of salbutamol, a single-blind crossover study in 10 subjects with stable asthma was designed. Each subject inhaled two puffs of salbutamol (200 μg) 1 hour after premedication with an aerosol preparation of either furosemide or placebo. Forced expiratory volume in 1 second (FEV 1 ) was measured before premedication, again before inhalation of salbutamol, and again 30, 60, and 90 minutes after, and 2, 3, 4, and 5 hours after inhalation of salbutamol. The influence of several phosphodiesterase-blocking agents on some mechanical and metabolic parameters of isolated perfused guinea pig hearts was studied in order to reinvestigate the repeatedly demonstrated close correlation between these functions. 1. In concentrations which caused 50% inhibition of the phosphodiesterase, theophyllin (1.8×10−4M), ethacrynic acid (4×10−4M) and papaverine (1×10−5M) produced positive inotropic effects by increasing the rate of contraction of the heart muscle (dp/dt). 1. The loop diuretic furosemide is known to antagonize the function of gamma-aminobutyric acid type A (GABAA) receptors. The purpose of the present study was to examine the direct interaction of furosemide with the GABAA receptors by autoradiography and ligand binding studies with native rat and human receptors and with recombinant receptors composed of rat subunits. 2. In this multicentre, double-blind trial, we randomly assigned normotensive patients with intermediate-risk PE to receive either a single 80 mg bolus of furosemide or a placebo.
