We investigated the role of GABA on intestinal motility using as model the murine distal colon. Effects induced by GABA receptors recruitment were examined in whole colonic segments and isolated circular muscle preparations to analyze their influence on peristaltic reflex and on spontaneous and neurally-evoked contractions. Using a modified Trendelenburg set-up, rhythmic peristaltic contractions were evoked by gradual distension of the colonic segments. Spontaneous and neurally-evoked mechanical activity of circular muscle strips were recorded in vitro as changes in isometric tension. The effects of GABA receptor agonists were investigated on guinea-pig isolated ileum longitudinal muscle with intact myenteric plexus. Electrical field stimulation (1 Hz, 10 s) of the histamine (1 microM)-precontracted preparation caused a contraction followed by a relaxation. Relaxations were inhibited by L-N(G)-nitroarginine (L-NA; EC50 3 microM) in a concentration-dependent manner. The inhibitory action of 10 microM L-NA was blocked by 10 microM L-arginine but not by D-arginine, which indicates that the relaxation was largely mediated by endogenous nitric oxide (NO). Focal cortical injuries are well known to cause changes in function and excitability of the surviving cortical areas but the cellular correlates of these physiological alterations are not fully understood. In the present study we employed a well established ex vivo-in vitro model of focal laser lesions in the rat visual cortex and we studied membrane and firing properties of the surviving layer 2/3 pyramidal neurons. Although an extensive body of literature confirmed γ-aminobutyric acid (GABA) as mediator within the enteric nervous system (ENS) controlling gastrointestinal (GI) function, the true significance of GABAergic signalling in the gut is still a matter of debate. GABAergic cells in the bowel include neuronal and endocrine-like cells, suggesting GABA as modulator of both motor and secretory GI activity. GABA effects in the GI tract depend on the activation of ionotropic GABAA and GABAC receptors and metabotropic GABAB receptors, resulting in a potential noteworthy regulation of both the excitatory and inhibitory signalling in the ENS. GABAb receptora agonista baklofena mazo devu atmiņu uzlabojošie efekti netransgēnā Alcheimera slimības modeļa žurkām

Alcheimera slimība ir neirodeģeneratīva saslimšana, kas ir biežākā no demences gadījumiem. Līdz šim esošie neiroprotektīvie preparāti nav spējuši apturēt neirodeģenerācijas procesus. Ir dati, ka gamma amino sviestskābes B tipa receptora (GABAB) agonists baklofens mazās devās uzlabo un protektē atmiņu. Šī pētījuma mērķis bija noskaidrot, vai ļoti mazas baklofena devas spēj uzlabot atmiņu Alcheimera slimības modeļa žurkām. Baklofens devās 0,025 un 0,05 mg/kg tika intraperitoneāli ievadīts kontroles grupas un Alcheimera slimības modeļa žurkām. Dzīvnieku atmiņa tika vērtēta ar Morris water maze testu.
