Characterization of Chlorolignins in Bleached Kraft Pulp Mill Effluents Using Elemental Analysis and Fingerprinting by CuO Oxidation And Hplc

Abstract Elemental analyses and chemical degradation techniques were applied to characterize lignin compounds in different stages of the chlorine bleaching process and in total mill effluent. Ultrafiltration was used for separation of chlorolignins into three nominal molecular weight fractions. Alkaline CuO oxidation products were analyzed by HPLC with diode array detection. Major compounds were identified by reference to authentic compounds. For comparison, native lignins (MWL), kraft lignins, and humic substances were studied as well. Parthenolide (PT) is a sesquiterpene lactone isolated from Tanacetum parthenium. In this study, PT showed varying cytotoxic effects against different solid tumor cell lines. HCT116 (p53+/+) colon carcinoma cells and their parental HCT116 knockout p53 (p53-/-) cell lines showed a resistance degree of 2.36. On the other hand, wild-type U87.MG cells or cells transfected with a deletion-activated EGFR cDNA (U87.MGΔEGFR) exhibited slight sensitivity toward PT. Multidrug-resistant MDA-MB-231-BCRP cells were even more sensitive toward PT than sensitive MDA-MB-231-pcDNA cells with a resistance degree of 0.07 (collateral sensitivity). Model compound synthesis for the structure determination of new unknown planar aromatic compounds originating from pulp mill

Abstract Several model compounds have been synthesized for verification of the structures of new unknown planar aromatic compounds (UPACs) observed in high-resolution gas chromatography/low-resolution mass spectrometry analyses of the polychlorodibenzodioxins (PCDDs) and polychlorodibenzofurans (PCDFs) in pulp mill products, effluents, sediments and mussels. The mass spectra of synthetic alkyl polychlorodibenzofurans (C6-PCDFs) were different, but those of alkyl polychlorobibenzyls (C5-PCBBs) were very similar compared to the spectra of UPACs.

Recent studies on extracellular RNA raised awareness that extracellular vesicles (EVs) isolated from cultured cells may co-purify RNAs derived from media supplements such as fetal bovine serum (FBS) confounding EV-associated RNA.
