G6PD Overexpression Protects Mice Against Associated Oxidative Stress and Delays the Occurrence of Frailty

To assess the impact of lifelong overexpression of G6PD on reactive oxygen species (ROS)-derived damage and the prevention of frailty, we measured the levels of macromolecular oxidative damage in young and old mice and the we tested the neuromuscular fitness and the grip strength in old mice. Old G6PD-Tg male and female mice showed diminished accumulation of DNA oxidation (measured as 8-hydroxyguanosine or 8-OHdG) in liver and brain. Old females also showed reduced lipid oxidation (measured as malondialdehyde or MDA) in the liver. Effects of strength and endurance training on antioxidant enzyme gene expression and activity in middle-aged men

This study was aimed at investigating the effects of a 21-week period of progressive strength or endurance training on peripheral blood mononuclear cells (PBMC) antioxidant enzyme gene expression and activity in healthy middle-aged untrained men. Field Desorption Mass Spectra of Gastrine Peptides and Glutathione Derivatives

Oligopeptides comprising the sequence of the C-terminal tetrapeptide of gastrine, Trp-Met-Asp-Phe-NH2, and several derivatives of glutathione, γ-Glu-Cys(SR)-Gly, were characterized by field desorption mass spectrometry. The field desorption mass spectra obtained at various field ion emitter temperatures reveal abundant molecular ions and fragmentation reactions that yield partial sequence information. In the series of glutathione derivatives investigated, characteristic ions formed by cleavage of the γ-Glu-Cys peptide bond determine the substituent at the Cys residue and can therefore be used to identify corresponding conjugation products of drug metabolites with glutathione.

Publisher Summary This chapter describes a method to accurately measure glutathione (GSH) and glutathione disulfide (GSSG) in the blood of humans and animals that perform physical exercise. With this method, it is possible to assess the oxidative stress associated with physical exercise by measuring changes in the blood GSH/GSSG ratio. Glutathione measurement is performed by a modification of the glutathione S -transferase method of Brigelius et al. This is based on the conjugation of chlorodinitrobenzene with GSH catalyzed by glutathione S -transferase. Cytoprotective effects of the antioxidant phytochemical indicaxanthin in beta-thalassemia red blood cells

Antioxidant phytochemicals are investigated as novel treatments for supportive therapy in beta-thalassemia. The dietary indicaxanthin was assessed for its protective effects on human beta-thalassemic RBCs submitted in vitro to oxidative haemolysis by cumene hydroperoxide. Indicaxanthin at 1.0-10 microM enhanced the resistance to haemolysis dose-dependently. In addition, it prevented lipid and haemoglobin (Hb) oxidation, and retarded vitamin E and GSH depletion. Oxidative damage to erythrocytes in thalassaemia has been related to generation of free radicals by an excess of denaturated alpha- or beta-globin chains, intracellular iron overload and low concentration of normal haemoglobin (HGB).
