Prospects of in vivo 31P NMR method in glyphosate degradation studies in whole cell system

Abstract The degradation of the phosphonate herbicide glyphosate ( N -phosphonomethylglycine) by four taxonomically distinct microorganisms was studied in vivo in whole cell system using phosphorus nuclear magnetic spectroscopy ( 31 P NMR). The time-course of glyphosate metabolization in dense cell cultures was followed by means of 31 P NMR up to 21 days after the addition. The results obtained by this non-invasive way confirmed that the cells of Spirulina platensis and Streptomyces lusitanus biodegrade herbicide. Trois types de gestion d’interculture (Labour, travail superficiel (TCS) et semis direct avec glyphosate (SD)) ont été comparés sur des parcelles conduites sans labour depuis 17 ans dont les 7 dernières années en ACS sur le domaine de l’INRA de Dijon avant implantation d’un blé, sur un dispo-sitif en bandes répétées sur quatre parcelles. Among diverse organophosphorus compounds, whose residues are determined in various ecosystems, phosphonates possess special meaning. Regarding their biological activity, capability to chelating metal cations, and stability of direct carbon to phosphorus bond under physiological conditions, phosphonic compounds found many various applications. N-Pyridylaminomethylenebisphosphonic acids constitute a class of promising herbicides. Since their mode of action at the cellular level is still poorly understood, we studied the influence of N-pyridylaminomethylenebisphosphonic acids on plant growth, at the whole plant and undifferentiated tissue levels, using seedlings and cell suspension cultures of mono- and dicotyledonous species. These compounds exhibited strong herbicidal properties, being equipotent with the popular herbicide glyphosate. Since they also depressed buckweed anthocyanin biosynthesis, the shikimate pathway could represent a site of action of N-pyridylaminomethylenebisphosphonic acids.

BEHAVIOUR OF GLUFOSINATE ANDGLYPHOSATE IN COMMONRAGWEED:APPARENT PARADOXAND SURPRISES We examined the foliar epidermis of commonragweed (Ambrosia artemisiifolia L.) plants and studied the behaviour of the herbicides glufosinate and glyphosate in this species. Both leaf surfaces exhibited 3 types of trichomes, according to their size and shape. Epicuticular waxes appeared amorphous, which may explain high levels of spray retention. Glufosinate and glyphosate were rapidly and almost completely taken up by common ragweed leaves; half maximum uptake was nearly reached after 5 and 3 h, respectively.
