Heteroduplex mobility assay (HMA) and DNA sequencing were performed on Flavescence doree (FD) phytoplasma strains and related phytoplasmas belonging to the elm yellows group. Part of the ribosomal RNA gene operon and a nonribosomal DNA region were utilized for phylogenetic analyses. Two FD strains, FD92 and FD-D, detected in France and Italy, respectively, were identical in both DNA fragments, confirming previous results. Other FD strains were all very similar and most closely resembled ALY, an Italian alder phytoplasma. Moscato Cerletti, a rediscovered aromatic cultivar with oenological potential in warm and dry areas

Baron Antonio Mendola was devoted to the study of grapevine, applying ampelography and dabbling in crosses between cultivars in order to select new ones, of which Moscato Cerletti, obtained in 1869, was the most interesting. Grillo, one of the most important white cultivars in Sicily, was ascertained to be an offspring of Catarratto Comune and Zibibbo, the same parents which Mendola claimed he used to obtain Moscato Cerletti. The complete genome of a Tomato black ring virus isolate (TBRV-Mirs) (RNA1, 7,366 nt and RNA2, 4,640 nt) and the RNA2 sequences (4,437; 4,445; and 4,442 nts) of three Grapevine chrome mosaic virus isolates (GCMV-H6, -H15, and -H27) were determined. All RNAs contained a single open reading frame encoding polyproteins of 254 kDa (p1) and 149 kDa (p2) for TBRV-Mirs RNA1 and RNA2, respectively, and 146 kDa for GCMV RNA2. p1 of TBRV-Mirs showed the highest identity with TBRV-MJ (94 %), Beet ringspot virus (BRSV, 82 %), and Grapevine Anatolian ringspot virus (GARSV, 66 %), while p2 showed the highest identity with TBRV isolates MJ (89 %) and ED (85 %), followed by BRSV (65 %), GCMV (58 %), and GA…

The study is focused on the viticulture of the island of Lampedusa, located in the strait of Sicily 113 km from the African coast and 205 km from the Sicilian coasts. It is a study that has no previous recorded documentations. The research was conducted on the island to a various number of vineyard farmers in order to describe cultivation techniques and the grape germplasm utilized. Somatic embryogenesis of Vitis vinifera L. (cv Sugraone) from stigma and style culture

Somatic embryo and plant regeneration were induced from stigma and style culture of grapevine (Vitis vinifera L. cv. Sugraone). To obtain somatic embryogenesis, explants were cultured on Nitsch and Nitsch basal medium (NN) supplemented with 88 mM sucrose and various combinations of the auxin β-naphthoxyacetic acid (NOA, 0-10 μM) and the cytokinin 6-benzylaminopurine (BA, 0-9 μM). Identification of aroma compounds of Vitis vinifera L. flowers by SPME GC-MS analysis

Using a gas chromatographic method (GC-MSanalysis), it was possible to determine the volatile constituent of an odorous flower from Vitis vinifera varieties growing in Sicily. More than 50 compounds were identified and the technique allowed us to determine that sesquiterpenes, as well as monoterpenes such as limonene and cymene, were the principal components. The odour-profiles allowed us to distinguish between variety groups or even single varieties.

Epidemiological survey on 'Candidatus Phytoplasma solani' and putative vectors in Western Sicily

Bois noir (BN) associated with ‘Candidatus Phytoplasma solani’ is an important grapevine yellows disease widespread in Europe and Mediterranean regions. During a two years survey (2014-15) carried out in a 10-year-old vineyard located in San Giuseppe Jato (PA), symptoms of Bois noir were observed on cvs Chardonnay, Pinot Noir and Nero d’ Avola. Incidence of the disease increased over the two years, reaching values of about 34 % on cv Chardonnay. Ca. ‘P. solani’ was detected on symptomatic leaf samples from all cultivars with the highest percentage on cv Chardonnay followed by Pinot noir and Nero d’Avola.
