Pinch Technique: Theory and Applications

We review the theoretical foundations and the most important physical applications of the Pinch Technique (PT). This general method allows the construction of off-shell Green’s functions in non-Abelian gauge theories that are independent of the gauge-fixing parameter and satisfy ghost-free Ward identities. We first present the diagrammatic formulation of the technique in QCD, deriving, at one loop, the gauge independent gluon self-energy, quark–gluon vertex, and three-gluon vertex, together with their Abelian Ward identities. Wick Theorem for General Initial States

We present a compact and simplified proof of a generalized Wick theorem to calculate the Green's function of bosonic and fermionic systems in an arbitrary initial state. It is shown that the decomposition of the non-interacting $n$-particle Green's function is equivalent to solving a boundary problem for the Martin-Schwinger hierarchy; for non-correlated initial states a one-line proof of the standard Wick theorem is given. Our result leads to new self-energy diagrams and an elegant relation with those of the imaginary-time formalism is derived. The theorem is easy to use and can be combined with any ground-state numerical technique to calculate time-dependent properties.

Differential staining of mucin granules from epoxy resin sections by a phosphotungstic acid-methyl green procedure.

After treatment of epoxy resin semithin sections from glutaraldehyde fixed rat large intestine with 5% aqueous phosphotungstic acid (PTA), staining with unpurified 0.2% solutions of methyl green at 60 C for 5 min produces a color differentiation between mucin granules of goblet cells. Some mucin granules and the glycocalyx appear deep green while the remaining granules, luminal mucin and collagen fibers are pink. The known contamination of unpurified methyl green with crystal violet seems to be responsible for the pink staining reaction of the latter structures, which also present an orange-red fluorescence under green exciting light. In vivo detection of cytokeratin filament network breakdown in cells treated with the phosphatase inhibitor okadaic acid.

We have previously described vulva carcinoma-derived A-431 subclone AK13-1, which stably expresses fluorescently labeled cytokeratin filaments (CKFs). Time-lapse fluorescence microscopy of these cells permits the continuous monitoring of the dynamics of the CKF cytoskeleton in vivo. To study mechanisms and principles of CKF disassembly as it occurs, e.g., during mitosis and liver disease, we have treated cells with the phosphatase inhibitor okadaic acid (OA), which induces complete CKF network breakdown within 3–5 h without significantly affecting the organization of the actin- and tubulin-based cytofilaments.
