Gibberellic acid stimulates lipid metabolism in barley aleurone protoplasts

Abstract Changes in the lipid composition of barley aleurone protoplasts following incubation in the presence or absence of gibberellic acid (GA) were studied and correlated with cell development. Analysis of neutral lipids in protoplast extracts and purified lipid bodies provide evidence for the strong effect of GA on the mobilization of storage lipids. In vivo protoplast staining with the membrane marker N -phenyl-l-naphthylamine (NPN) and phospholipid determinations of protoplast extracts show an increase in membrane lipids during protoplast incubation. Effects of Ga3 and Ca2+ on barley aleurone protoplasts: a freeze-fracture study

Freeze-fracture electron microscopy was used to study changes in the endomembrane system of barley (Hordeum vulgare L. cv. Himalaya) aleurone protoplasts. Protoplasts were used for this study because their response to calcium and the plant hormone gibberellic acid (Ga3) can be monitored prior to rapid freezing of cells for electron microscopy. Protoplasts incubated in Ga3 plus Ca2+ secrete elevated levels of a-amylase relative to cells incubated in Ga3 or Ca2+ alone. The endoplasmic reticulum (ER) and Golgi apparatus of protoplasts incubated in Ga3 plus Ca2+ undergo changes that are well correlated with the synthesis and secretion of a-amylase. Two field trials were carried out in Sicily during fall-winter and spring season to determine the influence of 9 fertilization treatments, obtained by changing the time (pre-plant and/or post-plant fertilization), and method (broadcast fertilization and/or fertigation) of fertilizer application and the type of fertilizer (mineral, organic, slow release fertilizers) on yield and quality of fennel. In the present study, the chemical compositions of the essential oils from roots, stems, leaves and fruits of Foeniculum vulgare subsp. piperitum collected in Sicily were evaluated by GC and GC-MS. The main components of the roots were terpinolene (33.15%), gamma-terpinene (12.18%) and fenchyl acetate (11.23%). Abstract Spot form of net blotch (SFNB), caused by Pyrenophora teres f. maculata is a prevalent and damaging foliar disease of barley (Hordeum vulgare). Fungicides are commonly used to manage SFNB infection and reduce production loss in susceptible varieties. The present work deals with the toxigenic mycobiota occurring in Spanish malting barley and the capability for producing mycotoxins by several important toxigenic fungi. One hundred and eighty seven samples of malting barley were gathered from Spanish breweries before processing. One hundred and fifty kernels per sample were surface-sanitized with a 2% sodium hypochlorite solution and incubated on three culture media. The most abundant fungi were species of Alternaria, Aspergillus, Penicillium and Fusarium, which were present in 93%, 82.3%, 57.8% and 27.8% of the samples, respectively.
