Preparation of extracts from mature spruce needles for enzymatic analyses

It was possible to extract simultaneously several active enzymes involved in the carbohydrate or the amino acid metabolism from spruce needles [Picea abies (L.) Karst.] when a) a 100 mM Na-Pi buffer of pH 7.5 containing 5% PVPP and 0.5% Triton X-100 was used and when b) the resulting crude extracts were freed from lowmolecular-weight compounds by gel-chromatography using the separation medium Fractogel TSK HW-40. Besides Triton X-100, Triton X-305, Myrij-52 and Brij-35 were tested, but 0.5% Triton X-100 brought about the most active enzyme extracts. The synthesis of a vinyl polymer with two different fluorescent dye end groups using reversible addition-fragmentation chain transfer (RAFT) polymerization is described. Use of a pentafluorophenyl (PFP) activated ester chain transfer agent (CTA) provided a polymer with an R end group that was reactive toward amines and a dithioester ω end group. The R PFP ester was amidated with Oregon Green Cadaverin. This did not harm the ω dithioester, which was subsequently aminolyzed with an excess of n-propylamine in the presence of Texas Red-2-sulfonamidoethyl methanethiosulfonate, resulting in a disulfide bond connecting the second dye to the polymer chain. A synthetic water-soluble meso-tetra(2,6-dichloro-3-sulfonatophenyl)porphyrinate of iron(III) chloride, Fe(TDCPPS)Cl, was employed as a biomimetic catalyst in the oxidative coupling of terrestrial humic materials. The synthesis of branched polyethylenes by ethylene polymerization with new tandem catalyst systems consisting of methylaluminoxane-preactivated linked cyclopentadienyl-amido titanium catalysts [Ti(η5:η1-C5Me4SiMe2NR)Cl2 (R = Me or tBu)] supported on pyridylethylsilane-modified silica (PySTiNMe and PySTiNtBu) and homogeneous dibromo nickel catalyst having a pyridyl-2,6-diisopropylphenylimine ligand (PyminNiBr2) in the presence of modified methylaluminoxane was investigated. Molecular weights of polyethylene (PE) obtained with using of zirconocene catalyst activated with large excess of methylaluminoxane (MAO) were determined by gel permeation chromatography (GPC) method. It was found that repeatability of the measurements is strongly influenced by MAO remained in PE samples as well as time of sample keeping in the apparatus (dissolution + measurement time) at temp. 142 °C. A method of MAO removing from PE samples, before GPC measurement, based on additional washing of the sample with HF in methanol solution has been proposed.
