Biopreservation by sugar and/or polymeric matrixes is a thoroughly studied research topic with wide technological relevance. Ternary amorphous systems containing both saccharides and proteins are extensively exploited to model the in vivo biopreservation process. With the aim of disentangling the effect of saccharides and polypeptidic crowders (such as gelatin) on the preservation of a model protein, we present here a combined differential scanning calorimetry and UV-vis spectrophotometry study on samples of myoglobin embedded in amorphous gelatin and trehalose + gelatin matrixes at different hydrations, and compare them with amorphous myoglobin-only and myoglobin-Trehalose samples. Introduction: Gel dosimeters for three-dimensional mapping of radiotherapy doses were introduced at Yale University in the mid-1980’s. Soon after, research and development in this field also started in Italy. Early work was done at the Istituto Superiore di Sanità and at the Universities of Pisa and Milan. Several institutes now collaborate on this topic with the goal of developing new formulations of hydrogel matrices with improved characteristics of stability, sensitivity and spatial resolution compared to those of earlier Fricke-gel and polymer-gel systems. Background Fast and effective treatment of hemorrhagic shock is one of the most important preclinical trauma care tasks e.g., in combat casualties in avoiding severe end-organ damage or death. In scenarios without immediate availability of blood products, alternate regimens of fluid resuscitation represent the only possibility of maintaining sufficient circulation and regaining adequate end-organ oxygen supply.
