1,4 dihidropiridinski derivati povećavaju ekspresiju gena Psma3, Psmb5 i Psmc6 u glasničkoj RNA štakora
2021
The ubiquitin-proteasome system modifies different cellular and protein functions. Its dysregulation may lead to disrupted proteostasis associated with multiple pathologies and aging. Pharmacological regulation of proteasome functions is already an important part of the treatment of several diseases. 1,4-dihydropyridine (1,4-DHP) derivatives possess different pharmacological activities, including antiaging and neuroprotective. The aim of this study was to investigate the effects of several 1,4-DHP derivatives on mRNA expression levels of proteasomal genes Psma3, Psmb5, and Psmc6 in several organs of rats. Rats were treated with metcarbatone, etcarbatone, glutapyrone, styrylcarbatone, AV-153…
A luminal glycoprotein drives dose-dependent diameter expansion of the Drosophila melanogaster hindgut tube
2012
An important step in epithelial organ development is size maturation of the organ lumen to attain correct dimensions. Here we show that the regulated expression of Tenectin (Tnc) is critical to shape the Drosophila melanogaster hindgut tube. Tnc is a secreted protein that fills the embryonic hindgut lumen during tube diameter expansion. Inside the lumen, Tnc contributes to detectable O-Glycans and forms a dense striated matrix. Loss of tnc causes a narrow hindgut tube, while Tnc over-expression drives tube dilation in a dose-dependent manner. Cellular analyses show that luminal accumulation of Tnc causes an increase in inner and outer tube diameter, and cell flattening within the tube wall,…
Gold nanoprobe-based detection of human telomerase reverse transcriptase (hTERT) Gene Expression
2015
Human Telomerase Reverse Transcriptase (hTERT) gene is expressed in all types of cancers, and it is considered as unique biomarker for early detection, monitoring and prognosis of different cancers. Routinely, the main techniques for detection of hTERT gene expression are based on enzymatic amplifications which need specified equipments, expert personnel and high cost and time. With regarding to the clinical importance of analysis of hTERT gene expression, we have developed a rapid, simple and low cost method which detects hTERT RNA target in 5 $\mu {\rm l}$ reaction scale using gold nanoprobes. The method is based on the inhibition of nanoparticle aggregation in the presence of ${\rm MgCl}…
Prolonging in utero-like oxygenation after birth diminishes oxidative stress in the lung and brain of mice pups☆
2013
Background Fetal-to-neonatal transition is associated with oxidative stress. In preterm infants, immaturity of the antioxidant system favours supplemental oxygen-derived morbidity and mortality. Objectives To assess if prolonging in utero-like oxygenation during the fetal-to-neonatal transition limits oxidative stress in the lung and brain, improving postnatal adaptation of mice pups. Material and methods Inspiratory oxygen fraction (FiO2) in pregnant mice was reduced from 21% (room air) to 14% (hypoxia) 8–12 h prior to delivery and reset to 21% 6–8 h after birth. The control group was kept at 21% during the procedure. Reduced (GSH) and oxidized (GSSG) glutathione and its precursors [γ-glut…
Induction of Apoptosis and Chemosensitization by the Histone Deacetylase Inhibitor Trichostatin in Hepatocellular Carcinoma Cells: Molecular Analysis…
2011
The mRNA and protein levels of RKIP are reduced and those of YY1 increased in clinical HCC. Loss, mutation, or promoter hypermethylation of the RKIP gene may not account for the downregulation of RKIP in HCC. Histone deacetylation can silence gene expression and play a significant role in hepatocarcinogenesis. The histone deacetylase inhibitor (HDACI) trichostatin induced cell growth inhibition and proapoptotic effects in HA22T/VGH and HepG2 HCC cells; it also exhibited synergy with doxorubicin. Treatment with trichostatin caused histone hyperacetylation and down- or upregulated expression of different genes (such as β-catenin, cyclin D1, hTERT, XIAP, and IL-6). These changes might, at leas…
The Ferroxidase Hephaestin in Lung Cancer: Pathological Significance and Prognostic Value
2021
AbstractIron is a fundamental nutrient utilized by living cells to support several key cellular processes. Despite its paramount role to sustain cell survival, excess of labile iron availability can inflict severe cell damage via reactive oxygen species generation which, in turn, can promote neoplastic transformation. The lung is particularly sensitive to iron-induced oxidative stress, given the high oxygen tensions herein present. Moreover, cigarette smoke as well as air pollution particulate can function as vehicles of iron supply, leading to an iron dysregulation condition shown to be crucial in the pathogenesis of several respiratory diseases including lung cancer. Hephaestin (HEPH) bel…
Global gene expression profiles in skeletal muscle of monozygotic female twins discordant for hormone replacement therapy
2010
Aging is accompanied by inexorable loss of muscle tissue. One of the underlying causes for this is the massive change in the hormonal milieu of the body. The role of a female sex steroid – estrogen – in these processes is frequently neglected, although the rapid decline in its production coincides with a steep deterioration in muscle performance. We recruited 54- to 62-year-old monozygotic female twin pairs discordant for postmenopausal hormone replacement therapy (HRT, n = 11 pairs; HRT use 7.3 ± 3.7 years) from the Finnish Twin Cohort to investigate the association of long-term, estrogen-based HRT with skeletal muscle transcriptome. Pathway analysis of muscle transcript profiles revealed …
Focusing on Ciona intestinalis (Tunicata) innate immune system. Evolutionary implications
2009
Phylogenetic analyses based on molecular data provide compelling evidence that ascidians are of critical importance for studying chordate immune system evolution. The Ciona intestinalis draft genome sequence allows searches for phylogenetic relationships, gene cloning and expression of immunorelevant molecules. Acidians lack of the pivotal components of the vertebrate recombinatory adaptive immunity, i.e., MHC, TCRs and dimeric immunoglobulins. However, bioinformatic sequence analyses recognized genic elements indicating the essential features of the Ig superfamily and ancestor proto-MHC genes, suggesting a primitive pre-duplication and pre-recombination status. C. intestinalis genes for in…
RNA Sequencing of Human Peripheral Blood Cells Indicates Upregulation of Immune-Related Genes in Huntington's Disease
2020
Huntington's disease (HD) is an autosomal dominantly inherited neurodegenerative disorder caused by a trinucleotide repeat expansion in the Huntingtin gene. As disease-modifying therapies for HD are being developed, peripheral blood cells may be used to indicate disease progression and to monitor treatment response. In order to investigate whether gene expression changes can be found in the blood of individuals with HD that distinguish them from healthy controls, we performed transcriptome analysis by next-generation sequencing (RNA-seq). We detected a gene expression signature consistent with dysregulation of immune-related functions and inflammatory response in peripheral blood from HD ca…
Iron Induces Proliferation and Morphogenesis in Primmorphs from the Marine SpongeSuberites domuncula
2002
Dissociated cells from marine demosponges retain their proliferation capacity if they are allowed to form special aggregates, the primmorphs. On the basis of incorporation studies and septin gene expression, we show that Fe3+ ions are required for the proliferation of cells in primmorphs from Suberites domuncula. In parallel, Fe3+ induced the expression of ferritin and strongly stimulated the synthesis of spicules. This result is supported by the finding that the enzymatic activity of silicatein, converting organosilicon to silicic acid, depends on Fe3+. Moreover, the expression of a scavenger receptor molecule, possibly involved in the morphology of spicules, depends on the presence of Fe3…
