Conventional methods based on hybridization and those based on biotechnology (somatic hybridization, genetic transformation, ploidy manipulation, etc.) are used to create novel genetic variations. We have carried out restoring of some Latvian alfalfa genetic resources from seeds of accessions that did not germinate in soil at all. Two of them were repatriated from the N. Vavilov All-Russian Institute of Plant Industry (multiplied more than 40 years ago) and three from the Research Institute of Agriculture (Latvia) (multiplied 15-20 years ago). For germination of old seeds we used early elaborated in vitro culture conditions. Germination rate ranged 2-60%, depending from the genotype and seeds storage conditions. An adequate management of the reproductive material for reforestation activities is fundamental in the conservation of forest genetic resources. In the present work the levels of genetic diversity and differentiation between artificial stands of Quercus ilex and the adjacent natural stands are assessed with nuclear microsatellites. Also, it was possible to analyse the natural population used as seed source for one of the reforestations studied.
