Implications of cysteine metabolism in the heavy metal response in Trichoderma harzianum and in three Fusarium species

We studied the ability of four different fungal species, Trichoderma harzianum, Fusarium antophyllum, Fusarium compactum and Fusarium phyllophilum, to grow in the presence of heavy metals, and monitored their cysteine and glutathione content and the activity of O-acetylserine(thiol)lyase (OASTL), which is involved in cysteine biosynthesis. Zn and Pb did not affect fungal growth or sporulation at the concentrations used, whereas Cd and Hg did. In most cases, cysteine and glutathione content was higher when fungi were grown in the presence of toxic metals. The use of glyphosate as the sole source of phosphorus or carbon for the selection of soil-borne fungal strains capable to degrade this herbicide

Abstract The herbicide glyphosate was used as a selection agent for isolation of fungal strains capable to degrade phosphorus-to-carbon bond from standard sandy-clay soil. The studies have shown that the herbicide used in Martin medium as a sole source of phosphorus br carbon caused the decrease of the fungal population and substantially changed strain composition, thus selecting those which are able to degrade glyphosate.

Abstract The interaction between the mycotoxin beauvericin (BEA) and 9 yeast strains of Saccharomyces cerevisiae named LO9, YE-2, YE5, YE-6, YE-4, A34, A17, A42 and A08 was studied. The biological degradations were carried out under aerobic conditions in the liquid medium of Potato Dextrose Broth (PDB) at 25 °C for 48 h and in a food/feed system composed of corn flour at 37 °C for 3 days, respectively. Emerging mycotoxins, such as enniatins and beauvericin, are common contaminants in vegetal matrices, but recently, the occurrence of mycotoxins in foodstuffs from animal origin has been also reported as they can be present in edible tissues of animals fed with contaminated feedstuffs. The active substance of new herbicide generation is N-(2-pirydyloamino) methylenobisphosphonic acid. Poly(oxycarbonylethylene) (poly-β-propiolactone), poly(oxycarbonyl-1,1-dimeothylethylene) (poly-β-pivalolactone), and poly(oxycarbonyltetramethylene) (poly-δ-valerolactone) were pyrolyzed directly in the ion source of a mass spectrometer and their thermal and electron impact induced degradation mechanisms are established. Poly-β-propiolactone is thermally degraded by rupture of the OCH2-bond by a cis elimination to give carboxyl and acryloyl endgroups, whereas poly-δ-valerolactone depolymerizes to δ-valerolactone. Poly-β-pivalolactone degrades thermally to cyclic oligomers of pivalolactone, which could be proved by a separated pyrolysis and IR-spectroscopy and gel chromatography.

International audience; The plasmid pADP1::Tn5 derived from pADP1[Atr(+)] carrying a TnS transposon conferring kanamycin and streptomycin resistances was constructed and introduced in Agrobacterium tumefaciens St96-4. This genetically modified strain was inoculated (similar to 108 cfu g(-1)) in potted soils planted with maize and treated or not with atrazine (1.5 mg kg(-1)). Bulk and maize rhizosphere soils were sampled 39 days after planting to look for soil indigenous bacteria that had acquired pADP1::Tn5. Aims: To investigate the role of horizontal gene transfer of mcd (methylcarbamate-degrading) gene in high genetic diversity of carbofuran-degrading bacteria.
