Soft X-ray Tomography Reveals HSV-1-Induced Remodeling of Human B Cells.

2022

Upon infection, viruses hijack the cell machinery and remodel host cell structures to utilize them for viral proliferation. Since viruses are about a thousand times smaller than their host cells, imaging virus-host interactions at high spatial resolution is like looking for a needle in a haystack. Scouting gross cellular changes with fluorescent microscopy is only possible for well-established viruses, where fluorescent tagging is developed. Soft X-ray tomography (SXT) offers 3D imaging of entire cells without the need for chemical fixation or labeling. Here, we use full-rotation SXT to visualize entire human B cells infected by the herpes simplex virus 1 (HSV-1). We have mapped the temporo…

Why viruses sometimes disperse in groups?

2019

AbstractMany organisms disperse in groups, yet this process is understudied in viruses. Recent work, however, has uncovered different types of collective infectious units, all of which lead to the joint delivery of multiple viral genome copies to target cells, favoring co-infections. Collective spread of viruses can occur through widely different mechanisms, including virion aggregation driven by specific extracellular components, cloaking inside lipid vesicles, encasement in protein matrices, or binding to cell surfaces. Cell-to-cell viral spread, which allows the transmission of individual virions in a confined environment, is yet another mode of clustered virus dissemination. Nevertheles…

Quantitative microscopy reveals stepwise alteration of chromatin structure during herpesvirus infection

2019

During lytic herpes simplex virus 1 (HSV-1) infection, the expansion of the viral replication compartments leads to an enrichment of the host chromatin in the peripheral nucleoplasm. We have shown previously that HSV-1 infection induces the formation of channels through the compacted peripheral chromatin. Here, we used three-dimensional confocal and expansion microscopy, soft X-ray tomography, electron microscopy, and random walk simulations to analyze the kinetics of host chromatin redistribution and capsid localization relative to their egress site at the nuclear envelope. Our data demonstrated a gradual increase in chromatin marginalization, and the kinetics of chromatin smoothening arou…

The article develops a reflection on a research experience gained within the University of Palermo in the context of the PO FERS “Fake News” project, which aimed to develop new and more effective automa…

Shooting borders. Per una geogafia visuale delle migrazioni

2017

Our perceptions of events in the Mediterranean are strongly influenced by a range of visual practices that impact on both the emotional and the political. In this paper, I set out to critically explore the visual geographies underlying photographic images of contemporary migration flows across the Mediterranean Sea. I argue the need for a methodological shift from a content analysis approach to a critical investigation of the spatial interweaving of images, gazes and aesthetic regimes that governs both the production and reception of such photographs. Given these theoretical premises, I focus on three images of migrants being rescued in the Mediterranean to illustrate how photography can ac…

Visual Methodologies and Geography's education in the pandemic time: notes on geopolitics of migration in the "Mediterranean Complex"

2022

<p>The relationship between Geography and the Visual has always been strong intertwined. As it is true that Geography has always operates through images (in the form of pictures, creative representations and above all cartographies), in the last two years, with the distance learning due to the spread of the Covid-19 pandemic, this phenomenon has not only increased, but it also became necessary. "The classroom as the most radical space", in the words of bell hooks, had to turn into a virtual space, where images have a fundamental role in the teaching/learning process. This paper wants to analyse the relationship between Geography and the Visual by analysing three image…

Explicit behavioral detection of visual changes develops without their implicit neurophysiological detectability

2011

Change blindness is a failure of reporting major changes across consecutive images if separated, e.g., by a brief blank interval. Successful change detection across interrupts requires focal attention to the changes. However, findings of implicit detection of visual changes during change blindness have raised the question of whether the implicit mode is necessary for development of the explicit mode. To this end, we recorded the visual mismatch negativity (vMMN) of the event-related potentials (ERPs) of the brain, an index of implicit pre-attentive visual change detection, in adult humans performing an oddball-variant of change blindness flicker task. Images of 500 ms in duration were prese…

2016

Daily mobility data are multidimensional and desaggregated, what make them hard to manipulate and to analyze. Although methods of representation and analyze have been proposed, none of them seem to be able to efficiently bring out knowledge from data. We propose to combinate this solutions and a methodology of innovative reaggregation to visualize and explore mobility data.
